重金属铬离子对胎鼠及仔鼠发育的影响

摘    要:目的探讨重金属铬离子对孕ICR小鼠胚胎以及仔鼠的发育影响,为行人工全髋关节表面置换术后重金属铬离子对妊娠机体可能产生的影响提供实验依据。方法将56只雌性受孕ICR小鼠随机分成高、中、低剂量组及对照组4组,每组14只。实验组以不同剂量重铬酸钾灌胃染毒,对照组予双蒸水灌胃。36只于孕19 d处死,观测着床数、黄体数、胎窝总重、胎盘总重、活胎数、死胎数。检测另20只母鼠娩出的仔鼠活产数、雌雄比、体重、身长、尾长。42 d处死仔鼠,测定其血清铬和肝脏铬含量,取肝脏组织作病理学观察。结果随染毒剂量的增加,中、高剂量组着床总数与对照组相比呈减少趋势,高剂量组胚胎存活率低于对照组,胚胎死亡率高于对照组。高剂量组仔鼠血清、肝脏铬含量高于中、低剂量组,差异有统计学意义(P〈0.01),高剂量组仔鼠肝脏细胞内出现大颗粒脂滴、染色体浓聚、线粒体减少的变化。结论本实验提示低剂量铬对胎鼠、仔鼠发育无影响,高剂量金属铬离子对孕鼠胚胎及仔鼠的发育产生较显著的影响,在子代体内仍存在毒性效应。
