摘    要:目的:建立适用测定假性胆碱酯酶(BChE)活性的微量羟胺法。方法:酶促反应用人血清BChE,选择优化以氯化丁酰胆碱为底物测定BChE活性微量羟胺法的条件并进行了方法学评价。结果及结论:本法底物丁酰胆碱最适浓度为9mmolL-1,最佳反应时间为30min,最适比色波长为492nm,灵敏度是以乙酰胆碱做底物的微量羟胺法的2倍,呈色物质比5,5-二硫基双-2-硝基苯甲酸(DTNB)法中的稳定,重复性好,操作简单,试剂及酶用量少,适用于临床检验及实验研究中BChE活性测定

Abstract:Objective: To establish the hydroxylaminederived microcolorimetric assay for the determination of butyrylcholinesterase(BChE) activity. Methods: Human serum butyrylcholinesterase was used in the enzymatic reaction with butyrylcholine as the substrate. The procedures were optimized and well conditioned with commentary. Results and Conclusion: The optimal conditions adopted in this method were as follows: butyrylcholine 9 mmolL-1, reactivation period 30 min and the colorimetric wavelength 492 nm. The sensitivity is two times as high as that when acetylcholine bromide is used as the substrate. The developed color is more stable than that in the DTNB method. This method is reproducible, easy to manipulate and samplesaving. It can be widely used in clinical determination of BChE as well as in basic research.
