骨髓基质细胞移植对大鼠脊髓损伤后神经细胞凋亡的影响

摘    要:目的研究骨髓基质细胞(BMSCs)移植对大鼠脊髓损伤(SCI)后神经细胞凋亡的影响,探讨骨髓基质细胞移植治疗脊髓损伤的机制。方法取大鼠股骨和胫骨骨髓培养BMSCs并传代。参照Taoka's方法制作大鼠脊髓压迫损伤模型。脊髓损伤后30min,进行BMSCs或DMEM培养液损伤周围局部四点注射。在术后3、7和14d,应用TUNEL方法检测脊髓损伤周围神经细胞凋亡情况。结果脊髓损伤后,损伤区周围组织内TUNEL阳性细胞数量明显增加,阳性细胞分布于脊髓灰质和白质内,但以白质内神经细胞为主。与DMEM组大鼠相比,移植术后3、7和14d,BMSCs移植显著减少脊髓损伤周围区凋亡的神经细胞数目(P〈0.05)。结论脊髓损伤后,BMSCs移植能够抑制神经细胞凋亡。
