递质和非递质性氨基酸在脊髓缺血性继发损伤中的作用

摘    要:用6300黄金系列氨基酸自动分析仪对清醒兔脊髓缺血20min,再灌不同时间后测定L4脊髓组织中的兴奋氨基酸(谷氨酸,门冬氨酸),抑制性氨基酸(甘氨酸,r-氨基丁酸,牛黄酸,丙氨酸)和非递质性氨基酸(谷氨酰胺,苏氨酸,丝氨酸)的变化。缺血20min时各氨基酸无变化,谷氨酸,门冬氨酸分别在再灌36h和48h显著下降,甘氨酸在再灌2h和48h,丙氨酸在2h和24h,r-氨基丁酸在灌36h下降显著,牛黄酸

Abstract:To induce ischemia of the spinal cord, the abdominal aorta was occluded below the origin of the renal arteries for 20 minutes. The tissue content of excitatory(glutamic and aspartic acid ), inhibitory(glycine, GABA, taurine and alanine) and neutral(glutamine, threonine and serine) amino acids was determined in the 4th lumbar segment of the spinal cord before and certain intervals after the reperfusion of the cord. It was found that all the amino acids showed no significant changes during ischemia, glutamate and aspartate were markedly decreased in the 36th and 48th hour after reperfusion, and glycine was decreased in the 2nd and 48th hour, GABA in the 36th hour , and alanine in the 2nd and 24th hour after reperfusion respectively. But taurine was increased in the 24th and 36th hour, glutamine was decreased in the 36th and 48th hour and threonine and serine were not changed at all the time points. The occurrence of the abnormal changes of the amino acids was consistent to the development of the secondary ischemic damages of the spinal cord.The mechanism of the change of each amino acid was discussed.
