| 摘 要： | 培养人脐带静脉内皮细胞并制备内皮细胞膜，以内皮细胞膜为抗原，经酶联免疫吸附法（Ｍ－ＥＬＩＳＡ）检测系统性红斑狼疮患者血清抗内皮细胞抗体，并将结果与细胞ＥＬＩＳＡ作比较。实验表明，两方法符合率为９２％，抗内皮细胞抗体阳性率两法差异无显著性（Ｘ￣２＝０．０６，Ｐ＞０．０５），抗体活性呈明显正相关（ｒ＝０．５１，Ｐ＜０．０１），但Ｍ－ＥＬＩＳＡ可检测ＩｇＧ、ＩｇＭ两型抗体，能节省大量试剂及血清，省时省力，便于推广，也适于制作药盒。

| Abstract: | ndothelial cell membranes(ECM)were prepared with endothelial cell monolayerscuItured from human umbilical cord vein,and anti-endothelial cell antibodies(AECA)were detected in serum samples of patients with SLE using ECM ELISA,The results of the ECM ELISA were compared with that of the cellular ELISA.The experiments showed that the positive rates of AECA were not statistically dif-ferent and significant correlation was present in AECA titers detected by the ECM ELISA and the cellu-lar ELISA. While the ECM ELISA can detect AECA of the IgG and IgM isotypes,and can save reagent and time,this assay should permit more detailed characterization of AECA properties. |
