无名异冲剂在骨折愈合中对DNA和BMP含量影响的实验研究

摘    要:目的探讨无名异冲剂治疗骨折的作用机理。方法新西兰家兔55只造成双侧桡骨3mm缺损的骨折后随机分为用药组和对照组,于术后1、2、3、4、5周取骨痂制成石蜡切片,以Feulgen反应和免疫组化技术分别显示DNA和BMP,进行图像定量分析。结果在骨折愈合过程中,骨痂中骨折修复细胞DNA指数和骨痂局部BMP含量的总体变化趋势两组相同;用药组第1、2周骨痂DNA指数和BMP含量明显高于同期的对照组(P<001);用药组BMP高峰出现在第2周,对照组出现在第3周。结论无名剂冲剂能促进骨折修复细胞的增殖,增加成骨细胞的活性,诱导BMP合成,加速骨折愈合。

Abstract:Objective To explore the effect and mechanism of Wumingyi Chongji on the treatment of fracture. Methods 55 New Zealand rabbits were selected to make radial fractures bilaterally with 3mm defect,and divided randomly into two groups,the drug administration group and the control.The bone calluses were taken out 1,2,3,4,5 weeks after operation.The paraffin sections of them were stained with Feulgen reaction and immunohistochemical technique to manifest DNA and BMP,respectively,for quantitative image analysis. Results In the process of fracture healing,the tendency of the total changes in the DNA index of fracture repairing cells and in the local content of BMP in calluses was the same in both groups;1 and 2 weeks after operation,DNA index and BMP content in the calluses of drug administration group was significantly higher than that of the control(P<001);the peak value of BMP in drug administration group was appeared at the second week,and that in the control at the third week. Conclusion Wumingyi Chongji can promote the proliferation of fracture repairing cells in callus,increase the activity of osteoblast,induce the BMP synthesis and accelerate the union.
