摘    要:目的 比较两种矽肺造模方法的优劣,并探讨一次性气管滴注法造模的最佳时间。方法 口鼻暴露染尘小鼠20只,气管滴注染尘小鼠20只,分别于染尘后第7、14、21、28天处死;对照组小鼠5只,正常饲养28d处死。检测各时间节点小鼠肺湿重、脏器系数、肺组织羟脯氨酸(hydroxyproline,HYP)含量,并观察肺组织病理改变。结果 口鼻暴露法染尘21、28d小鼠肺湿重及脏器系数,7、14、21、28d小鼠肺纤维化评分,与对照组比较差异有统计学意义(P<0.05),染尘28d小鼠肺组织形成矽结节;气管滴注染尘后第14、21、28天小鼠肺湿重、脏器系数,第28天小鼠HYP含量,第7、14、21、28天小鼠肺纤维化评分,与对照组比较差异有统计学意义(P<0.05),染尘后第21天形成多个矽结节,第28天矽结节融合成大片状。结论 暴露法染尘28d小鼠开始形成矽结节,滴注法染尘后第21天小鼠即可完成矽肺造模。

Abstract:Objective Compare the advantages and disadvantages of two silicosis modeling methods and explore the optimal time for trachea.Methods Both 20 mice were exposed to dust by daily mouth/nose inhalation or tracheal instillation,then were killed on 7,14,21,28days after dust exposure;5 mice in control group were fed normally and killed on 28th day of experiment.Detect the lung wet weight, organ coefficient, hydroxyproline content in lung tissues, and pulmonary pathological changes of the rats at each time point.Results The results showed that compared with control group,the lung wet weight and organ coefficient at 21 day and 28 day,and the pulmonary fibrosis score at 7,14,21 and 28day of dust exposed mice by mouth/nose inhalation method had statistical significance(P<0.05);silicosis nodules in lung tissue were found at the 28th day after dust exposure.While the lung wet weight and organ coefficient of mice exposed dust by tracheal instillation method were found significantly different from control group as early as the 14th day of dust exposure,the significant differences of HYP content and pulmonary fibrosis score of mice were found significantly different from control group at the 28th day and the 7th,14th,21st,28th day(P<0.05),furthermore,the multiple silicosis nodules were formed at the 21th days of dust exposure, and on the 28th day,it was found that the silicosis nodules even fused into large patches.Conclusion The results suggested that only after 28 days dust-exposure,the silicosis nodules could begin to formed in mice by mouth/nose inhalation method,in the dust-exposed mice by tracheal instillation method,the silicosis modelling only need 21 days.
