摘    要:目的 :检查金雀异黄素(genistein,GEN)及其新型合成类似物7-二氟甲氧基-5,4'-二-正辛烷氧基金雀异黄素(7-difluoromethoxyl-5,4'-di-n-octyl genistein,DFOG)对人卵巢癌OVCAR3源性球细胞迁移和Twist1/FoxM1蛋白表达的作用.方法 :Transwell小室细胞迁移试验检测细胞迁移率.蛋白质印迹分析细胞Twist1和FoxM1蛋白表达水平.结果 :与OVCAR3细胞相比,OVCAR3源性球细胞迁移率更高.GEN(40μM)或DFOG(5μM和10μM)抑制OVCAR3源性球细胞迁移.此外,GEN(40μM)或DFOG(5μM和10μM)平行地下调Twist1和FoxM1蛋白表达.结论 :GEN及其类似物DFOG抑制OVCAR3源性球细胞体外迁移与其抑制Twist1和FoxM1蛋白表达相关.

Abstract:Objective To investigate the effects of genistein (GEN) and its novel synthetic analogue 7-difluoromethoxyl-5, 4'-di-n-octyl genistein (DFOG) on the cell migration and the expression of Twist1/FoxM1 in human ovarian cancer OVCAR3-derived spheroids. Methods Cell migration ratio was detected using Transwell cell migration assay. The expression of Twist1 and FoxM1 was analyzed using western blot. Results Compared with OVCAR3 parent cells, cell migration ratio was higher in OVCAR3-derived spheroids. Moreover, both GEN (40μM) and DFOG (5μM and10μM) could inhibited the cell migration ra-tio in OVCAR3-derived spheroids. And they could also parallelly down-regulate the protein expression of Twist1 and FoxM1. Conclusion Cell migration inhibited by GEN and its analogue DFOG is associated with inhibiting the expression of Twsit1 and FoxM1 in OVCAR3-derived spheroids.
