摘    要:目的使用二次回归正交旋转组合设计优化A群脑膜炎奈瑟氏菌培养基的配方。方法采用摇瓶培养方式,设计四因素二次回归正交旋转组合设计优化培养基配方,使用SASV9.1软件进行数据分析,并进行验证培养。结果优化后四因素最佳浓度为L-谷氨酸5班,水解酪蛋白15g/L,酵母浸膏15g/L,葡萄糖5班,使用优化培养基进行3次验证试验,A600平均值为2.278,与预测值2.299基本相符。结论使用二次回归旋转组合正交设计优化A群脑膜炎奈瑟氏茵培养基配方切实可行,效果理想。

Medium optimization of Neisseria meningitidis serogroup A by quadratic orthogonal rotation combination design

Abstract:Aim to optimize medium of Neisseria meningitidis serogroup A by quadratic orthogonal rotation combination design. Methods Using shake flasks for fermentation after design four factors quadratic orthogonal rotation combination to optimize culture formula,then the data was analysed by SAS V9.1 software. At last,3 tests verified for optimized formula. Results The optimum concentrations of 4 factors were L-glutamic acid 5g/L,casein 15g/L,yeast extract 15g/L,glucose 5g/L. 3 tests using optimized formula were verified, and the average value of A600 was 2.278, without apparent difference compared with the predicted value 2.299. Conclusion The quadratic orthogonal rotation combination design can be used to optimize the medium for Neisseria meningitidis serogroup A.
