嗜酸乳杆菌对生殖道上皮细胞的黏附和黏附拮抗作用的研究

摘    要:目的 比较热灭活状态和活菌状态乳酸杆菌对生殖道上皮细胞的黏附性及其对生殖道常见致病菌白色念珠菌和加德纳菌的黏附拮抗作用。方法 运用光镜和电镜分析嗜酸乳杆菌活菌和热灭活两种生物状态对刮取的阴道上皮细胞和传代培养的宫颈上皮细胞系HeLa细胞的黏附指数以及经两种生物状态嗜酸乳杆菌拮抗作用后致病菌的黏附指数的变化 ,并用台盼蓝染色法比较二者黏附HeLa细胞后对细胞存活率的影响。结果 两种生物状态的嗜酸乳杆菌对刮取的阴道上皮细胞的黏附不存在显著性差异 ,而热灭活状态对HeLa细胞的黏附指数显著高于活菌状态 ,两种生物状态的嗜酸乳杆菌对白色念珠菌和加德纳菌均有黏附抑制作用 ,热灭活状态嗜酸乳杆菌黏附HeLa细胞在一定时间内不降低细胞存活率。结论 经热灭活的嗜酸乳杆菌对生殖道上皮细胞有较强的黏附性并可对女性生殖道常见致病菌发挥黏附拮抗作用。

Abstract:Objective To compare the adhesive ability of the inactivated and living lactobacilli to the scraping human vaginal epithelial cells and HeLa cells and the antagonistic ability to uropathogens, Candida alibicans and Gardnerella vaginalis. Methods Microscope and Scanning electron microscope were used to analyze the adherence index of the lactobacilli in two different forms to the scraping vaginal epithelial cells and HeLa cells as well as the changes of the adherence index of the uropathogens after antagonism with the lactobacilli, the vitality of HeLa after the adherence of lactobacilli was observed by trypan blue assay. Results There was no significant difference in the adherence index of the lactobacilli in two different forms to the scraping vaginal epithelial cells, but the adherence index of the heat-killed lactobacilli was higher than that of the living ones, and they both had antagonistic ability to C.albicans and G.vaginalis. The vitality of HeLa did not decrease within certain period. Conclusion The inactivated lactobacilli had potential to adhere to the genital epithelium cells and could exert antagonistic effect to frequent uropathogens.
