平哮颗粒药效学及抗炎机理研究

摘    要:目的 评价平哮颗粒止咳、化痰、抗炎、平喘药理作用,探讨其抗炎作用机理.方法 通过氨水诱咳、气道酚红排泄、二甲苯致耳肿胀、乙酰胆碱诱喘,分别评价平哮颗粒的止咳、化痰、抗炎、平喘作用.比较血清IgE、IL-13、IFN-γ改变探讨平哮颗粒抗炎作用机理.结果 平哮颗粒高、低剂量组止咳作用均优于空白组(P<0.01),高剂量组与美敏伪麻组效果相当.平哮颗粒高、低剂量组化痰作用与氨溴索组无明显差异,均优于空白组(P<0.01).平哮颗粒高、低剂量组抑制耳肿胀作用不及泼尼松组,均优于空白组(P<0.01).平哮颗粒高剂量组平喘效果与氨茶碱组疗效相当,二者优于低剂量组(P<0.05),各组均优于模型组(P<0.01).平哮颗粒高剂量组在降低IgE、IL-13,升高IFN-γ方面与泼尼松相当,与模型组比较具有统计学差异(P<0.01).结论 平哮颗粒抗气道炎症的机制与抑制IgE生成,调节Th1/Th2免疫有关.

Abstract:Objective:To evaluate the pharmacological effect of Pingxiao granule regarding mechanisms of relieving cough,resolving phlegm,and soothing asthma and to explore its anti-inflammatory effects at the same time.Methods:Both high-dose and low-dose Pingxiao granule groups were included in each experiment,with corresponding western medicine group as control group.Ammonia induced cough,airway phenol red excretion,xylene induced ear swelling,and acetylcholine induced asthma were used to evaluate the effect of Pingxiao granule on mechanism of relieving cough,resolving phlegm,soothing asthma and resisting inflammatory respectively.Comparison the change of IgE,IL-13 and IFN-γ in rats serum to explore the anti-inflammatory action mechanism of Pingxiao Granule.Result:Either high-dose or low-dose Pingxiao Granule group showed better relieving cough effect than control group.The effect of high-dose group was similar to Huifeining group.High-dose and the lowdose Pingxiao granule groups showed similar effects in resolving phlegm with the ambroxol group.All of above groups had better results than control group.Neither high-dose nor low-dose Pingxiao granule group could match Perdnisone group on inhibiting ear swelling.But they are all better than that of control group.High-dose Pingxiao granule group had the similar effect with Aminophylline group on soothing asthma.The two groups were better than low-dose Pingxiao granule group.Any of them was better than control group.High-dose Pingxiao granule group was equivalent to Perdnisone group on decreasing IgE and IL-13,and increasing IFN-γ which had significant difference compared with model group.Conclusion:Pigxiao Granule has obvious anti-inflammatory,antitussive,antitussive,expectorant and antiasthmatic effects.Its mechanism of anti airway inflammation was related to inhibiting production of IgE and regulating Th1/Th2 immune system.
