盐酸戊乙奎醚对体外循环大鼠肝脏损伤的影响

摘    要:目的 探讨盐酸戊乙奎醚对体外循环大鼠肝脏损伤的影响.方法 健康清洁级成年雄性SD大鼠32只,随机分为4组(n=8),假手术组(Sham组)、模型组(Mod组)、盐酸戊乙奎醚低剂量组(L-PHC组)和高剂量组(H-PHC组).Sham组只置管不转流,Mod组体外循环预充液中不加入盐酸戊乙奎醚,L-PHC组和H-PHC组体外循环预充液中分别加入盐酸戊乙奎醚0.6mg/kg和2mg/kg.于停止体外循环即刻抽取左股动脉血2ml后处死,测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性,观察肝脏组织超微结构改变.结果 与Sham组比较,Mod组、L-PHC组、H-PHC组血清ALT,AST活性升高(P<0.05);与Mod组比较,L-PHC组、H-PHC组血清ALT、AST活性降低(P<0.01),病理损伤程度减轻;与L-PHC组比较,H-PHC组血清ALT、AST活性降低(P<0.05),病理损伤程度减轻.结论 盐酸戊乙奎醚0.6mg/ks和2mg/kg可减轻体外循环诱发大鼠肝脏损伤的程度,2mg/kg的效果更明显.

Effects of penehyclidine hydrochloride on liver injury induced by cardiopulmonary bypass in rats
Abstract:Objective To investigate the effects of penehyelidine HCl on liver injury induced by cardiopulmonary bypass(CPB)in rats.Methods Thirty-two healthy adult male SD rats weighing 300-450g were randomly divided into 4 groups (n=8 each):group I sham operation(s);groupⅡCPB;group Ⅲ low-dose penehyclidine+CPB (L-PHC) and group IV high-dose penehrclidine+CPB(H-PHC).The animals were anesthetized with intraperitoneal 10% chloral hydrate 0.3 ml/100g, intubated and mechanically ventilated.Femoral artery and right auricle (via jugular vein)were cannulated.CPB was performed for 60 min.Penehyrclidine Hel 0.6 mg/kg and 2 mg/kg were added to the priming solution in group Ⅲ and IV respectively.Blood samples were taken from femoral artery at he termination of CPB for determination of serum alamine aminotransferase (ALT) and aspartate aminotransferase (AST) and the animals were killed.The liver was removed for electron microscopic examination.Results Serum ALT and AST activities were significantly higher in group II,Ⅲ andⅣ than in group C and were significantly lower in group Ⅲ and IV than in groupⅡ.The electron microscopic examination showed that the histologic damage was significantly reduced on group Ⅲ and Ⅳ as compared with group Ⅱ,Penehyclidine HCl exerts protective effects againdst liver injury induced by CPB in.a dose-dependent manner.Conclusion Penehyclidine HCI(0.6 and 2.0 mg/kg) can reduce liver injury induced by CPB dose-dependently.
