二氧化硅粉尘对实验大鼠肺泡巨噬细胞的毒性效应

摘    要:目的观察二氧化硅粉尘对大鼠肺泡巨噬细胞(AMs)的毒性作用。方法测定不同二氧化硅(SiO2)粉尘浓度培养AMs24小时以及培养不同时间的AMs上清中乳酸脱氢酶(LDH)、丙二醛(MDA)含量,并记录不同浓度粉尘刺激AMs的死亡率。同时以二氧化钛粉尘为阴性对照。结果巨噬细胞死亡率随粉尘浓度的增加而增加,AMs上清中LDH及MDA含量与SiO2粉尘浓度呈良好的剂量-反应关系,且与其刺激时间呈良好的时间-效应关系。结论染SiO2粉尘的实验大鼠巨噬细胞呈现明显的毒性效应。

Abstract:Objective To observe the toxicity effect of Sprague-Dawley rat alveolar macrophages(AMs) exposed to silica dust.Methods Lactate dehydrogenase(LDH)and malondialdehyde(MDA)in supernatants of AMs exposed to different concentrations of silica dust and different periods of one concentration of silica dust were detected.The mortality rate of AMs exposed to different concentrations of silica dust was recorded.Those exposed to titanium dioxide dust(TiO2) were as negative control groups.Results The mortality rate of AMs increased with the increase of concentration of silicon dioxide(SiO2) dust.It was shown that there was good dose-effect and time-effect relationships between LDH,MDA levels and concentration of SiO2 and exposure time.Conclusion There wasa obvious toxicity effect on rat AMs when exposed to SiO2.
