胆脂瘤患者外耳道上皮细胞增殖和凋亡的研究

摘    要:目的 :检测胆脂瘤上皮及胆脂瘤患者外耳道上皮增殖细胞核抗原 ( PCNA)、Ki- 67抗原和凋亡细胞。方法 :取后天继发性中耳胆脂瘤患者和正常人外耳道皮肤 (各 5例 )及胆脂瘤上皮 ( 1 3例 )。应用免疫组化和原位切口末端标记 ( TUNUL)法检查 PCNA、Ki- 67抗原和凋亡细胞的分布和数量。结果 :胆脂瘤患者外耳道皮肤 PCNA、Ki- 67抗原的分布和数量与胆脂瘤上皮相似 ,而较正常外耳道上皮分布范围广 ,阳性细胞多且深染 ;凋亡细胞数量与正常外耳道上皮相比有明显增加趋势。结论 :外耳道上皮异常增殖和细胞凋亡可能与中耳胆脂瘤的发生有关

Abstract:Objective:To study on the relationship between the proliferation and apoptosis of auditory meatal skin in patients with cholesteatoma. Methods:Five samples of auditory meatal skin in cholesteatoma, 5 samples of normal auditory meatal skin and 13 samples of cholesteatoma epithelium were selected. Immunohistochemistry analysis and TUNEL method were used in the experiment. Results:A number of positive cells were observed in the basal and suprabasal layers in auditory meatal skin samples of cholesteatoma and in epithelium of cholesteatoma than those in normal auditory meatal skin. An increasing tendency of apoptotic cells was seen in the outmost layer of auditory meatal skin samples of cholesteatoma. Conclusion:The results surggested that the disturbance of proliferation and cell apoptosis in auditory meatal skin in patients suffering from cholesteatoma may play an important role in the pathogenesis of middle ear cholesteatoma.
