消黄方对α-萘异硫氰酸酯大鼠黄疸模型肝组织氧化应激的影响

摘    要:目的:探讨消黄方对α-萘异硫氰酸酯(ANIT)大鼠黄疸模型的治疗作用及抗氧化应激效应。方法:Wistar雄性大鼠40只,随机分为正常组(10只)和ANIT处理组(30只)。ANIT按照2mL/kg一次性经口灌胃,48h后,ANIT处理组随机分为模型组,消黄方组及熊去氧胆酸(UDCA)组(各10只),各组每天分别给予生理盐水、消黄方、UDCA治疗1周,实验结束处死全部大鼠。观测大鼠死亡率、检测肝功能及肝组织总超氧化物歧化酶(T-SOD)、谷胱甘肽-S转移酶(GST)活性和丙二醛(MDA)含量。结果:肝功能结果发现:与正常组比较,模型组大鼠肝ALT、AST、ALP、TBIL、DBIL显著升高(P0.05,P0.01)。与模型组比较,UDCA和消黄方显著降低血清TBIL和DBIL含量(P0.01),消黄方显著降低ALT、AST及ALP活性(P0.05,P0.01)。肝组织氧化应激结果显示:与正常组比较,模型组肝组织GST活性及MDA含量显著升高(P0.01),T-SOD活性显著下降(P0.01),而消黄方显著降低GST活性及MDA含量(P0.01),升高肝组织SOD活性(P0.01)。结论:消黄方具有保肝退黄作用;氧化应激是ANIT大鼠黄疸模型的重要病理环节,抗氧化应激效应是消黄方退黄主要机制之一。

Abstract:Objective: to investigate the anti-oxidative stress effects of Xiaohuang decoction(XHD) in ANIT-induced jaundice using UDCA as a positive control in rats.Methods: Male Wistar rats(40),were randomly divided into normal group(10) and ANIT-treated(30) group.After ANIT treated 48h,ANIT-treated group were randomly divided into model group,XHD group and UDCA group(10 in each).At the end of the experiment all rats were sacrificed.Liver function and total superoxide dismutase(T-SOD),glutathione-S transferase(GST) activity and malondialdehyde(MDA) content in liver tissue were determined.Results: Compared with normal rats: ALT,AST,ALP,TBIL,DBIL was significantly higher(P<0.05,P<0.01) in ANIT rats.XHD and UDCA reduced significantly TBIL and DBIL content(P<0.01).XHD,but not UDCA lowered significantly ALT,AST and ALP activity(P<0.05,P<0.01).Compared with normal group,GST activity and MDA levels increased significantly(P<0.01) and T-SOD activity was decreased significantly(P<0.01) in ANIT rats,while the XHD reduce GST activity and MDA content(P<0.01),elevated liver T-SOD activity(P<0.01).Conclusion: Anti-oxidative stress action should be one important mechanism for XHD in reducing jaundice in rats.
