斯氏肺吸虫成虫10—30KD抗原蛋白的分离及在免疫诊断中的应用

摘    要:在酶免疫转移印渍技术(EITB)分析斯氏肺吸虫虫体成份,显示10~30KD抗原蛋白(主带22、24和26KD)对该病具免疫诊断价值的基础上,将SDS-PAGE和电洗脱技术相结合,分离并纯化出斯氏肺吸虫成虫的10~30KD蛋白组份,采用Dot-ELISA方法免疫诊断肺吸虫。10~30kD的斯氏肺吸虫成虫抗原与斯氏、卫氏肺吸虫病人血清均呈阳性反应,与其它吸虫病和健康人血清末产生反应。该结果进一步表明10~30KD斯氏肺吸虫成虫抗原的Dot-ELISA为肺吸虫病高度特异、敏感的免疫诊断方法。并为两种肺吸虫的共同抗原,可用于免疫诊断斯氏、卫氏肺吸虫病。

Abstract:In our previous study, adult worm antigen of Pagumogonimus skrjabin (P. S) was an alyzed by Enzyme-immuno transier blotting (EITB), and its 10-30kD proteins (especially protein bands of 22. 24. 26kD) were shown to be recognizec immunologically by the sera of infected humans and animals. In the present experiment, the 10-30kD proteins from adult worms of P. S were separated and purified by slab SDS-PAGE and electroelution and their immunodiagnostic value was assessed by Dot-ELISA. The sera of 34 cases of paragonimiasis showed positive reaction in the recognition of 10-30kD antigens, while control sera, includng other trematodiasis and healthy persons, registered negative reaction without exception. The results indicated that Dot-ELISA of 10-30kD adult antigens was a higb sensitive and specific immunodiagnostic method, which could diagnoze pagumogonimisis (P. S) as well as paragonimisis (P. W).
