PLP-组氨酸脱羧酶的纯化与晶体制备

摘    要:本文成功地改进了依赖磷酸吡哆醛的组氨酸脱羧酶(PLP-Histidine Decarboxylase;PLP-HDCase)的纯化方法。通过采用离子交换层析,减少浓缩及透析次数,方法简便。同时,增加Aminohexyl-sepharose层析柱床体积,使一次获得纯化PLP-HDCase量增加10~15倍。在获得足够优质PLP-HDCase的基础上,找到了稳定结晶及晶体生长条件,首次获得大单晶。

Abstract:This paper described a modified method of purifying the Morganella pyridoxal-5-phosphate histidine decarboxylase (PLP-HDCase). The yield of PLP-HDCase once increased 10 to 15 times by using only DEAE-cellulose column and Aminohexyl-sepherose column, and the time of purification was reduced. Besides by obtaining sufficient stable crystal condition big crystal was established. Asa result , a big crystal was obtained for the first time.
