摘    要:目的测定铁皮石斛鲜品中氨基酸的含量。方法样品以异硫氰酸苯酯(PITC)为衍生化试剂衍生后进行HPLC分析。采用CAPCELLPAKC18SG300(250ram×4.6mm,51a,m)色谱柱,柱温为250(:;流动相A为0.05mol/L醋酸钠缓冲液(以醋酸调pH值至6.5)-乙腈(1:1),流动相B为0.05mol/L醋酸钠缓冲液(以醋酸调pH值至6.5),梯度洗脱,流速为0.6ml/rain;检测波长为254nm。结果15种氨基酸浓度在0.077-2.042μg范围内呈良好的线性关系(r:0.9994-0.9999),平均回收率为94.3%-107.7%。结论本方法灵敏、准确,具有良好的重复性和稳定性,可用于铁皮石斛中氨基酸的检测。

Abstract:Objective To determine and analyze amino acids in Dendrobium officinale Fresh Product. Methods The sample was derivatized with phenyi isothiocyanate ( PITC). HPLC was per - formed on a CAPCELL PAK Clg SG300 ( 4.6ram x 250ram, 5 μm) column with gradient elution of 0, 05mol/L sodium acetate buffer solution ( adjusted to pH 6.5 with acetic acid) - acetonitrile ( 1: I ) (A) and 0.05mol/L sodium acetate buffer solution( adjusted to pH 6.5 with acetic acid) (B) at the flow rate of 0.6ml/min,deteeted at 254nm. Column temperature was 25~C. Results Fifteen aminoacids had good linearity in the ranges of 0. 077 -2. 042p, g (r :0. 9994 -0. 9999).The average recoveries were 94. 3% - 107. 7%. Conclusion The method is sensitive and accurate with good stability,which is useful for the determination of amino acids in Dendrobium officinale.
