苦瓜提纯液对糖尿病肾病大鼠肾脏MMP-9、TIMP-1 mRNA表达的影响

摘    要:目的探讨苦瓜对糖尿病肾病大鼠细胞外基质及基质金属蛋白酶9(MMP-9)、组织抑制剂1(TIMP-1)基因表达的影响。方法将60例Wistar雄性大鼠随机分为对照组、模型组、苦瓜组及洛汀新组,采用链脲佐菌素一次性腹腔内注射法诱导糖尿病模型,造模1、5周测定各组24h尿蛋白、肾功能及血糖,5周后取各组肾组织用RT—PCR技术检测MMP-9、TIMP-1的基因表达。结果模型组较对照组肾小球MMP-9、TIMP-1 mRNA表达增加,反映肾功能的各项生化指标水平升高。苦瓜组、洛汀新组较模型组MMP-9、TIMP-1 mRNA表达减少,肾功能有所改善。结论苦瓜对糖尿病大鼠肾结构功能具有保护作用,除降低血糖外,调节肾小球MMP-9、TIMP-1表达从而减轻肾小球细胞外基质沉积也可能是其作用途径之一。

Abstract:Objective:To explore the effects of charantia on the gene expression of extracellular matrix(ECM),matrix metalloproteinase-9(MMP-9) and tissu inhibitors of metalloproteinase-1(TIMP-1) mRNA diabetic Nephropathy rats.Methods:60 Wistar rats were randomly divided into the normal control gruop,the model group,the charantia treatment group and the benazepril treatment group.The murine models with DN induced by STZ.24h urinary albmuen and Glu,Scr,BUN in serum were determined every week.MMP-9 and its main inhibitor tissue inhibitor of TIMP-1 mRNA was analyzed RT-PCR five weeks after DN rats modeled.Results:BUN,Ccr,and Upro in diabetic rats were significant higher than that in model group.Compared with those in kidneys of model group,diabetic kindeys showed increased expressions of both MMP-9,TIMP-1 mRNA excessive deposition of glomeruli extracellular matrix.The level of expressions of MMP-9,TIMP-1 mRNA were reduced by charantia treatment,renal function was improved.Conclusion:Charantia has some renal protective effection diabetic enphropathy partly through inhibition of excessive deposition of glomeruli extracellular matrix by down-regulating MMP-9 and TIMP-1.
