摘    要:目的:优化HPLC测定人血清中万古霉素浓度的方法.方法:在不同流动相配比、沉淀剂及柱温条件下,高效液相色谱法测定血万古霉素浓度,确定最佳的色谱条件和操作方法.结果:在色谱条件为色谱柱C18(250.0 mm×4.6 mm,5μm)、流动相磷酸二氢钾缓冲液(pH 3.2)-乙腈(90:10,v/v)、流速1.0 ml/min、检测波长236 nm、柱温35℃的情况下,万古霉素浓度在4.47~60.83 mg/L范围内线性关系良好(r=0.9 943,n=5),回收率在85%~115%范围内;日内、日间RSD<15%,在定量下限附近RSD<20%.结论:优化后,操作简便快速,回收率稳定,适于临床血药浓度测定.

The improved monitoring and determination for serum vancomycin concentration

Abstract:Objective: To optimize the method for the determination of serum vancomycin concentra tion by HPLC. Methods: Chromatographic peaks of serum vancomycin concentration under differ- ent mobile phase, precipitant and column temperature were analyzed to find the best cbromato graphic condition and operation method. Results: The better linearity range of vancomycin concen- tration was between 4.47 mg/L and 60.83 mg/L(r=0. 9 943, n=5)by using the hypersil C18 (250.0 mm×4.6 mm,5μm), potassium dihydrogen phosphate(pH3. 2)-acetonitrile (90 : 10, v/ v) as mobile phase, flow rate 1.0 ml/min, detection wavelength 236 nm and column temperature 35℃, and the recovery rate was at therange of 85% to 115%. The RSD values of intra-day and in ter-day were lower than 15%, and the RSD values of LLQ lower limit of quantification were lower than 20 %. Conclusion: The improved method is simple and rapid, and the recovery rate is stable It is suitable for the application of clinical and scientific research.
