牛磺酸对压力超负荷大鼠心肌中钙调神经磷酸酶和肾上腺髓质素的影响

摘    要:目的观察牛磺酸灌服6周后对压力超负荷大鼠心肌中钙调神经磷酸酶(CaN)和肾上腺髓质素(ADM)的影响。方法将Wistar大鼠随机分为3组:对照组、缩窄组和给药组。参照Anderson方法制作压力超负荷心肌肥厚大鼠模型。术后1周开始给予牛磺酸50mg·kg^-1·d^-1,给药6周后处死动物,检测左心系数(左室重/体重比,LVW/BW)、CaN活性及ADM含量。结果与对照组相比,缩窄组大鼠SBP、LVW/BW显著升高(P〈0.05),心肌中CaN活性、ADM含量显著增高(P〈O.05)。给药组与缩窄组相比,SBP、LVW/BW下降(P〈0.05),心肌中CaN活性、ADM含量显著下降(P〈0.05)。结论牛磺酸有降低压力超负荷大鼠收缩压及减轻心肌肥厚的作用,该作用可能与其抑制CaN活性和ADM分泌有关。

Abstract:Objective To observe the influence of taurine to calcineurin (CaN) and adrenomedullin (ADM) in the myocardium of pressure overload rats which was treated with 6 weeks of taurine. Methods The Wistar rats were randomly divided into 3 groups: sham-operated group, partial-operated group, taurine-treated group. Referring to the Anderson method, manufacture the model of pressure overload rats, a week later, taurinetreated group was given taurine by 50 mg·kg^-1·d^-1. After been treated by six weeks, the systolic blood pressure (SBP) of big mouse was measured, left ventricu|ar mess index (LVW/BW) was calculated, the activity of CaN and the level of AMD were measured. Results Comparing with sham-operates group, the SBP and LVW/BW of partial-treated groups were increased (P〈0.05), the activity of CaN and the level AMD in cardiac muscle were increased (P〈0.05). Comparing with partial-operated groups, SBP and LVW/BW of taurine-treated group were decreased (P〈0.05), and the activity of CaN and the level AMD were decreased (P〈0.05). Conclusion Taurine can decrease SBP and produce a good preventive effect on the pressure overload rats by decreasing the activity of CaN and the level of ADM.
