复方苦参注射液对人子宫内膜癌细胞HEC-1B雌激素受体表达的影响

摘    要:目的探讨复方苦参注射液对人子宫内膜癌细胞(HEC-1B)雌激素受体(ER)亚型ER-α36、ERα-66的影响,探讨其作用机制。方法 HEC-1B细胞给予高、中、低3个浓度复方苦参注射液、苦参注射液、白土苓注射液、顺铂注射液处理,采用四甲基偶氮唑蓝(MTT)法观察各给药组对HEC-1B细胞增殖的影响。HEC-1B细胞给予中浓度复方苦参注射液、苦参注射液、白土苓注射液、顺铂注射液处理,采用反转录聚合酶链反应法(RT-PCR)观察不同给药组对HEC-1B细胞雌激素受体ER-α36、ERα-66表达水平的影响。结果与空白对照组比较,高浓度复方苦参注射液、苦参注射液均显著抑制HEC-1B细胞增殖(P〈0.05)。复方苦参注射液、苦参注射液、顺铂注射液作用于HEC-1B细胞48 h后,与空白对照组比较,细胞中ER-α36 mRNA表达水平均明显降低(P〈0.05);除白土苓组外,其他各给药组ERα-66mRNA表达水平亦明显降低(P〈0.05)。结论复方苦参注射液抑制HEC-1B细胞增殖的作用机制可能与抑制细胞雌激素受体ER-α36、ER-α66的基因表达有关。

Abstract:Objective To study the effect of Fufangkushen injection on the expression of estrogen receptor(ER)-α36 and ER-α66 in endometrial cancer cell line HEC-1B,and explore its mechanism.Methods HEC-1B cell proliferation was measured by MTT method,which was treated by high-,medium-,low-dose of Fufangkushen injection,Kushen injection,Baituling injection and positive drug DDP respectively.The expressions of ER-α36 and ER-α66 were analyzed by using RT-PCR method after treated for 48 h with medium-dose of drug respectively.Results HEC-1B cell proliferation was significantly inhibited after 48 h or 72 h treatments by high concentration of Fufangkushen injection and Kushen injection compared with the control group(P<0.05).The mRNA expressions of ER-α36 and ER-α66 were reduced significantly after 48 h treatments of Fufangkushen injection,Kushen injection and DDP(P<0.05).Conclusion The anticancer effect of Fufangkushen and Kushen injection might be related with reducing mRNA expressions of ER-α36 and ER-α66 in endometrial cancer cell line HEC-1B.
