半边莲煎剂对小鼠H22肝癌荷瘤细胞系C-erbB-2和P53表达的影响

摘    要:目的观察半边莲煎剂对实验性小鼠H22型肝癌的抑制作用。方法小鼠右腋下皮下种瘤造成实体瘤模型,昆明种小鼠24只(♀♂各半),随机分为3组,即中药组、西药组、对照组,每组8只。造模后分别给予半边莲煎剂灌胃、5-FU腹腔注射、生理盐水灌胃给药,连续给药14d。第15天以颈椎脱臼法处死,并将肿瘤组织取出称其质量,计算抑瘤率;制作小鼠肿瘤细胞切片,用免疫组织化学方法对肿瘤细胞切片进行染色,检测半边莲煎剂对肿瘤的抑制作用及对C-erbB-2和P53表达的影响。结果半边莲煎剂对小鼠肝癌细胞的生长抑制率为33.98%,C-erbB-2及P53在3组中均呈阳性表达,中药组C-erbB-2表达减弱,P53表达升高,与对照组比较差异有统计学意义(P均〈0.05)。结论半边莲煎剂对小鼠H22型肝癌有明显的抑制作用,其机制可能与肿瘤细胞内C-erbB-2、P53蛋白表达有关。

Abstract:AIM To observe the inhibitive effects of lobelia ehinensis lour decoction on tumor-bearing mice. METHODS After injected H22 hepatoma eells into their right axial, 24 Kunming mice, half male and half female were randomly and equally divided into 3 groups: traditional Chinese medicine group, 5-Fu group and control group. Mice of control group were given normal saline by intragastrie administration. The mice in Chinese medicine group were given lobelia ehinensis lour decoction by intragastrie administration, and those of western medicine group were injected with 5-Fu. All animals were fed for 14 d continuously. On the 15th day all the rniee were killed by cervical vertebra disloeative whose tumors were taked out and weighted. Then the antitumor rate was ealeulated, and sliees were made for immunohis- tochemical analysis on C-erbB-2 and P53 expression. RESULTS The inhibitive rate was 33.98 % in lobelia ehinensis lour decoction treated group and that of 5-Fu was 50.48%. The expression of C-erbB-2 was signifieantly decreased and the expression of P53 was significantly increased in traditional Chinese medicine group. CONCLUSION The inhibitive effects of lobelia ehinensis lour decoction on H22 cells are significant and the mechanism may be related with the expression of C-erbB-2 and P53.
