去氢钩藤碱对谷氨酸功能低下精神分裂症模型小鼠自主活动和探究行为的影响

摘    要:目的:观察不同剂量去氢钩藤碱对MK-801所致的谷氨酸功能低下小鼠模型自主活动及探究行为的影响。方法昆明种小鼠66只分组,进行下述对照观察:①取30只为A组:观察去氢钩藤碱高、中、低剂量(2.6、1.3、0.75mg/kg)灌胃3d对昆明种小鼠自主活动和探究行为的影响;②取36只为B组:给予浓度为0.072mg/mLMK-801腹腔注射诱导小鼠自主活动增加,观察去氢钩藤碱各组对MK-801所致小鼠高活动性的影响。结果①A组:去氢钩藤碱各组与空白对照组比较,活动总路程差别不明显(P〉0.05)。②B组:模型组与空白组比较活动总路程明显增加(18692.47±4108.67 vs 6893.08±1210.11,P〈0.01),去氢钩藤碱各剂量组与模型组相比:活动总路程明显减少(9736.95±870.22,9396.5±1460.13,9416.05±1266.25vs18692.47±4108.67,P〈0.05);中央区和周边区活动路程也有不同程度减少(P〈0.05)。结论①去氢钩藤碱对正常小鼠自主活动和探究行为无影响;②去氢钩藤碱对MK-801所致的谷氨酸功能低下小鼠自主活动和探究行为的增高有下调节作用。

Abstract:Objective To observe the different doses of Dehydrogenase rhynchophylline on MK-801-induced mouse model of glutamate dysfunction locomotor activity and exploration behavior, to further explore the pharmacological effects of dehydroepiandrosterone Rhy. Methods 66 Kunming mice of clean grade divided into two parts(n=6)for the following processes:①Take 30 for the group A:control group (pure water gavage), positive drug group (amisulpride solution gavage), dehydroepiandrosterone Rhy high, medium and low dose group 2.6, 1.3, 0.75mg/kg)concentration solution gavage for three consecutive days, to observe the effect of above treatment in Kunming mice on locomotor activity and exploration behavior;②Take 36 for the group B:control group and model group(pure water gavage), positive drug group(amisulpride solution gavage), dehydroepiandrosterone Rhy high, medium and low dose group (2.6, 1.3, 0.75mg/kg)concentration solution gavage, intragastrically three days, intraperitoneal injection of saline half-hour after the intragastric, the other group to 0.36mg/kg body weight MK-801 intraperitoneal injection induced locomotor activity increase in mice, observed in each group for the the impact of high activity induced by MK-801 in mice. Results①A Group:dehydroepiandrosterone rhynchophylline each group compared with the control group, no signiifcant difference in total distance events (P〉0.05).②B groups:model group compared with the control group signiifcantly increased total distance events (18692.47 ± 4108.67 vs 6893.08 ± 1210.11, P〈0.01), dehydroepiandrosterone Rhy each dose group compared with model group:Active Total distance was signiifcantly reduced (9736.95±870.22,9396.5±1460.13, 9416.05 ± 1266.25vs18692.47 ± 4108.67, P〈0.05);central area and the surrounding area there are different degrees of activity from reduced (P〈0.05). Conclusion ①Dehydrogenase rhynchophylline had no effect in normal locomotor activity and exploratory behavior in mice.②Dehydrogenase Rhy can reduce the locomotor activity and exploratory behavior increased by MK-801-induced in mice.
