摘    要:目的建立测定麻黄附子细辛汤水煎液中麻黄碱含量(以盐酸麻黄碱计)的反相高效液相色谱法。方法色谱柱:Diamonsil-C18柱(200mm×4.6mm);流动相:乙腈:水(含0.02mol·μL^-1磷酸二氢钾和0.2%三乙胺)(10:90);流速:1.0mL·min^-1;检测波长:210nm;柱温:27℃。结果样品中盐酸麻黄碱的色谱峰与其他组分完全达到基线分离,回归方程为:y=2229.4854x-22.2885,r=0.9998(n=6),线性范围0.1776—1.3320μg,平均加样回收率为101.45%(RSD=I.9%,n=6)。结论所建立的测定方法快速简便,重现性良好,可用于麻黄附子细辛汤水提物中麻黄碱的含量测定。

Abstract:OBJECTIVE To establish a RP-HPLC method for the determination of ephedrine(determined as ephedrine hydrochloride)in the water decoction of mahuang-fuzi-xixin-soup. METHODS The sample was separated on a Diamonsil-C18(200mm × 4.6mm)with the mobile phase consisting of acetonitrile and water(containing 0.02mol·μL^-1 potassium dihydrogen phosphate and 0.2 % triethy lamine) ( 10 : 90) at a flow rate of 1.0mL·min^-1 The detection wavelength was 210nm, and the column was kept at 27℃. RESULTS Chromatographic separation of ephedrine hydrochloride and the co-existed compounds achieved baseline resolution, y = 2229. 4854x-22.2885, r = 0. 9998 (n = 6), the calibration was linear in the range of 0.1776-1.3320μg, anti the average recovery of ephedrine hydrochloride was 101.45% (RSD= 1.9%, n = 6) .CONCLUSION The method is rapid, simple and reproducible and can be used for the quantity detection of ephedrine in the water decoction of Mahuang-Fuzi- Xixin-soup.
