急性脑梗死患者中性粒细胞活化的研究

摘    要:目的探讨中性粒细胞活化在急性脑梗死疾病患者中的意义。方法抽取急性血栓形成性脑梗死患者及健康志愿者外周血,运用流式细胞技术检测中性粒细胞表面黏附分子CD62L和CD 11b/CD18表达。运用放射免疫分析检测血浆IL-6、IL-8、TNF-α水平,用酶联免疫吸附试验检测血浆IL-10水平。结果相对于正常组,脑梗死组中性粒细胞CD11b/CD18平均阳性表达率显著升高(P〈0.001);CD 62L平均阳性表达率明显降低;细胞因子水平均明显升高(P〈0.001)。结论在急性血栓形成性脑梗死的急性期,中性粒细胞处于活化状态。以细胞黏附为表现的中性粒细胞活化加快了血栓的进程,可能是血栓形成的重要发病原因之一。

Abstract:Objective To study with acute cerebral infarction. Methods the significance of polymorphonuclear neutrophil activation in patients Forty never treated cerebral infarction (CI) subjects have been selected to the study, thirty healthy subjects were studied as control groups. Expression of CD 62L and CD 11b/ CD 18 on PMN were analyzed by flow eytometry(FCM). Radioimmuno assay(RIA) were used to detect plasma level of IL-6, IL-8, and TNF-α, Enzyme-linked immunosorbent assay (ELISA) were used to measure plasma level of IL-10. Results The average positive expression percentage of CD 11 b/CD 18 of neutrophil from the patients with acute CI is higher than the control group(P 〈0.01 ) , and th.e average positive expression percentage of CD 62L is lower than the control group(P〈0.01). The levels of plasma cytokines in the CI groups were increased markedly compared with normal control (P〈 0. 001 ). Conclusion Expression of CD 11b/CD 18 and CD 62L and level of plasma eytokines might be regarded as the marker of PMN activa- tion, PMN activation plays an important role in acute CI by expedite the progress of thrombosis, increase the inflammation around thrombus and degree of injured blood vessel.
