摘    要:目的:通过高效液相色谱(HPLC)研究了灯盏花乙素苷元的离解常数和油水分配系数。方法:利用待测组分的保留因子(k值)与流动相的pH值,导出了pH-k的曲线方程,并通过曲线拟合软件求出拐点即是灯盏花乙素苷元的离解常数。采用振荡-液相色谱法测定了灯盏花乙素苷元的油水分配系数。结果∶HPLC对灯盏花乙素苷元离解常数的测定中k-pH曲线符合S型数理模型,灯盏花乙素苷元的油水分配系数在水相pH3.0~8.0范围内不随pH变化而变化。结论∶用所建立的方法分别测定了灯盏花乙素苷元的离解常数和油水分配系数,HPLC法准确度高,重现性好,方便简单,分析快速、准确。

Abstract:Objective: To study the dissociation constant and oil/water partition coefficient of scutellarein by pH-chromatography.Methods: The dissociation constant of scutellarein is calculated respectively from the retention factors-pH of mobile phase in the pH-chromatography;A oscillate method was established to determine n-octanol/water partition coefficient of scutellarein.Results: The curve of pH-chromatography showed to be suitable for Logistic fit;The pH 3.0-8.0 of buffer solution had no influence on the distribution coefficient of scutellarein.Conclusions: pH-chromatography can be used to determine the dissociation constant of scutellarein accurately;The oscillate method can be used to determine the oil/water partition coefficient of scutellarein.
