摘 要: | 目的:探讨与肝丹主治功能有关的药理药效学作用及其特点。方法:2.2.15细胞株模型及改进方法;大鼠CCl4慢性肝损伤模型,小鼠腹腔巨噬细胞吞噬功能及血清溶血含量实验,结果;动态观察13d肝丹对HB-sAg,HBeAg的最大抑制率为76.47%(d7)71.43%(d5),且呈现HBV分泌,复制越是旺盛,抑制作用越强的特点;

PHARMACODYNAMIC EXPERIMENTAL STUDY ON TREATMENT OF CHRONIC HEPATITIS BY GANDAN |

Abstract: | OBJECTIVE:Gandan is a mixture of several Chinese herbs. This study is to assess the therapeutic effects of Gandan on several experimental models. METHODS: 2.2.15 cell line was used to observe the inhibiting action of Gandan by detecting HBsAg, HBeAg and HBV-DNA once daily for 13 days. The protective effect of Gandan on chronic hepatic injury induced by CCl4 was assessed.The effect of Gandan on immune function was observed by in mouse macrophage and antibody for sheep RBC in mouse serum phagocytosis. RESULTS :Gandan showed inhibiting action on HBsAg and HBeAg secretion and HBV-DNA replication in 2.2.15 cell line, the highest inhibiting rates to HBsAg and HBeAg were 76.47% (on day 7) and 71.43% (on day 5) respectively. The inhibiting action of Gandan was in a dose-dependent manner. In chronic CCl4 hepatic injury model, Gandan could significantly reduce the injury of liver tissue as comparing with the model of control group [ALT (U/L):39.80±7.91,72.00±17.42; AST (U/L):32.20±3.55,53.90±11.35 ] and inhibited liver fibrosis. Gandan could increase the number of peritoneal macrophage in mouse, and also increased the number of antibody to sheep RBC in mouse serum. CONCLUSION: Gandan is effective in inhibiting replication or proliferation of HBV in 2.2.15 cells, reducing liver injury in CC14 intoxicated rats and regulating immune reaction of the host, so Gandan would be effective therapy in treatment of chronic hepatitis B. |
