祛风通络方对微小病变型肾病大鼠P-selectin的作用

目的：探讨P-selectin在微小病变型肾病大鼠中的表达及祛风通络方对其的干预作用。方法：56只雄性清洁级SD健康大鼠随机分为空白对照组（A组）和AN模型组,采用一次性尾静脉注射阿霉素（ADR）的方法,建立大鼠AN模型,造模成功后再将模型组分为病理组（B组）、祛风通络方组（C组）、泼尼松组（D组）和贝那普利组（E组）,邻苯三酚红/钼酸盐法测定24h尿蛋白量;电子显微镜观察肾小球超微结构;自动生化分析仪测定血生化指标;ELISA法测定P-selectin浓度。结果：模型组大鼠于实验第21天时尿蛋白排泄较正常对照组明显增高（P〈0.01）;药物干预后,各药物组大鼠尿蛋白排泄均显著下降,与病理组比较（P〈0.01）;祛风通络方组比模型组大鼠肾小球基底膜足突数目增多、且排列较整齐;与A组比较,血清Alb明显下降（P〈0.01）,血清TG、TC、Scr及P-selectin均显著升高（P〈0.01）;与病理组比较,祛风通络方组血生化指标及P-selectin均有一定程度的改善（P〈0.05或P〈0.01）。结论：P-selectin可能参与了微小病变型肾病的发病过程,祛风通络方可能通过降低P-selectin的表达从而对肾脏起到保护作用。

Effects of Qufeng Tongluo Recipe on P-selectin of Minimal Change Nephropathy Rats

Objective：To investigate the expression of P-selectin in Serum of minimal change nephropathy rats and effects of QuFeng TongLuo Recipe（QFTL） on it.Methods：56 Sprague-Dawley（SD） rats were divided at random into normal control（A） and AN model groups.The model rats were established by single transvenous injection of Adriamycin and the rats were subdivided into model（B）,QFTL recipe（C）,prednisone（D）,and benazepril（E）groups 3 weeks later.Then 24 h urinary protein were count,glomerular ultramicrostructure were detected by electron microscope,biochemical indexes were detected by automatic biochemistry analyzer,and the expression of P-selectin in Serum were detected by ELISA.Results：Compared with group A,the model group showed significantly increased level of 24 h urinary protein（P0.01）.Treatment with QFTL recipe obviously lowered the 24 h urinary protein（P0.01） and increased the foot process counts on the glomerular filtration membrane in the AN rats.Compared with group A,serum albumin level decreased obviously（P0.01） while serum TC、TG and P-selectin levels increased（P0.01）.Treatment with QFTL recipe obviously improved biochemical indexes and P-selectin in the AN rats（P0.01）.Conclusion：P-selectin may play an important role in the progression of minimal change nephropathy.QFTL recipe has renal protective effect in minimal change nephropathy rats,partially through down-regulating the expression of P-selectin.
