蛇六谷乙酸乙酯萃取物A_1组分对大鼠胶质瘤C_6细胞增殖及C-myc、bcl-2、p~(53)癌基因表达的影响

摘    要:目的:探讨蛇六谷乙酸乙酯萃取物A1组分对大鼠胶质瘤C6细胞增殖的抑制作用及其作用机制。方法:用MTT比色法检测不同浓度A1组分对C6细胞增殖的抑制作用,RT-PCR法检测A1组分对C6细胞C-myc、bc l-2,p53mRNA基因表达的影响。结果:A1组分对C6细胞的增殖具有明显的抑制作用,且呈量-效关系,并有明显的细胞形态学改变;RT-PCR结果显示,随着A1组分作用剂量的增加,C6细胞的C-myc、bc l-2及p53mRNA表达均有明显降低。结论:蛇六谷乙酸乙酯萃取物A1组分对大鼠胶质瘤C6细胞的增殖具有明显的抑制作用,其作用机制可能与抑制原癌基因C-myc、bc l-2及p53的表达有关。
