益肾胶囊对糖尿病肾病大鼠足细胞podocin表达的影响

目的：观察益肾胶囊对糖尿病肾病（DN）大鼠足细胞Podocin表达的影响。方法：采用切除右侧肾脏后腹腔一次性注射STZ的方法建立DN模型,实验分组为：正常对照组、DN模型组、益肾胶囊组、贝那普利组,每组各10只,益肾胶囊组每只大鼠灌胃益肾胶囊625g·kg-1·d-1,贝那普利组每只大鼠灌胃贝那普利10mg·kg-1·d-1,正常对照组及DN模型组每日给予等量的蒸溜水。经过治疗12周后,采用PT-PCR及Westernblot检测肾组织中Podocin表达的情况。结果：实验12周末,DN模型组足细胞Podocin蛋白表达明显低子正常对照组（P〈0.05）。经过益肾胶囊治疗后Podocin表达明显增加,与DN模型组比较（P〈0.05）,与贝那普利组比较两治疗组之间差异无统计学意义。结论：益肾胶囊明显改善DN模型大鼠足细胞Podocin的表达,可能具有一定的足细胞损伤保护作用。

Effect of Yishen Capsule on Podocin in Renal Tissue of Rats with Diabetic Nephropathy

Objective：to investigate effect of yishen capsule on podocin in renal Tissue of Rats with diabetic nephropathy.Methods：40 male wistar rats were randomly divided into four groups：control group,diabetic nephropathic model group,yishen capsule group and Benazepril group for 12 weeks,each group have10 rats.Then rats of model groups have been treated by streptozotocin（STZ） after there right kidney had been excised.The evels of blood glucose were examined at the end of 2,4,6,8,12 weeks.the expression of podocin were examed by RT-PCR,western blot and immunofluorescence.Results：At the end of 12 weeks,the expression levels of podocin was lower than control group（P0.05）.after treatment of yishen capsule.The expression levels of podocin were higher than those in diabetic nephropathic model group（P0.05）.Conclusion：yishen capsule could regulate the expression of Podocin,which may have protect effects on podocytes on renal tissue of rats with diabetic nephropathy.
