电针心包经穴对MCAO大鼠NR2A、NR2B表达的影响

摘    要:目的:探讨电针心包经穴对MCAO大鼠不同时相点NR2A、NR2B表达的影响。方法:采用线栓法制备大鼠大脑中动脉栓塞(MCAO)模型,将其随机分为电针心包经组和模型组,再将每1组二次随机分为1d、3d、7d、14d、21d5个亚组。对电针心包经组进行电针治疗,用Western blot检测NR2A、NR2B的蛋白表达,用行为学评分评估大鼠处理前后运动功能障碍与恢复情况。结果:①行为学评分:同1时相点与处理前比较,模型组21d亚组处理后评分降低;电针心包经组3d、7d、14d、21d亚组处理后评分均明显降低,差异均有统计学意义。②NR2A、NR2B蛋白表达:同1时相点与模型组相比,电针心包经组1d、3d、7d亚组的NR2A、NR2B表达均明显降低,而14d、21d亚组的NR2A、NR2B表达均显著上升,差异均有统计学意义。结论:电针治疗脑梗死的作用机制可能与对不同时相点NR2A、NR2B的表达进行差异性调节,从而提高缺血耐受性,保护受损的神经细胞有关。

Abstract:Objective:To investigate the effect of electroacupuncture at the points of the pericardium meridian on the expression of NR2A and NR2B at different time points in MCAO rats.Methods:The middle cerebral artery embolism (MCAO) model of rats was prepared by the thread embolization method,and they were randomly divided into electroacupuncture pericardial meridian group and model group,and each group was randomly divided into 1d,3d,7d,14d,21d5 subgroups.Electroacupuncture was performed on the electroacupuncture pericardium meridian group.Western blot was used to detect the protein expression of NR2A and NR2B,and the behavioral score was used to evaluate the motor dysfunction and recovery of rats before and after treatment.Results:①Behavioral score:Compared with the pre-treatment at the same time point,the scores of the 21d subgroup of the model group were reduced after treatment;the scores of the electroacupuncture pericardial meridian group 3d,7d,14d,and 21d subgroups were significantly reduced after treatment,and the differences were all There is statistical significance.② Protein expression of NR2A and NR2B:Compared with the model group at the same time point,the expressions of NR2A and NR2B in the subgroups 1d,3d,and 7d of the electroacupuncture pericardial meridian group were significantly reduced,while the expressions of NR2A and NR2B in the subgroups 14d and 21d were significantly reduced Both increased significantly,and the differences were statistically significant.Conclusion:The mechanism of electroacupuncture in the treatment of cerebral infarction may be related to the differential regulation of the expression of NR2A and NR2B at different time points,thereby improving ischemic tolerance and protecting damaged nerve cells.
