不同浓度米非司酮对体外培养子宫肌瘤细胞周期的影响及其诱导的细胞凋亡

摘    要:目的:研究不同浓度米非司酮对体外培养子宫肌瘤细胞凋亡的影响。方法:采用体外培养的子宫肌瘤细胞,用不同浓度米非司酮处理后培养48h,用流式细胞仪分析子宫肌瘤细胞周期和凋亡率的改变。结果:不同浓度的米非司酮均可诱导子宫肌瘤细胞的凋亡,随着米非司酮浓度的增加,子宫肌瘤细胞的凋亡率有增高的趋势(P<0·05),低浓度米非司酮(10-8~10-7mol/L)可使G2/M期子宫肌瘤细胞比例减少(P<0·05),高浓度米非司酮(10-6~10-4mol/L)可使S期细胞比例减少(P<0·05)和G2/M期细胞比例减少(P<0·01)。结论:米非司酮可引起体外培养子宫肌瘤细胞的凋亡,呈现浓度-效应关系,低浓度米非司酮和高浓度米非司酮可能引起不同时期子宫肌瘤细胞的凋亡。

Abstract:Objective:To study the effects of different concentrations of mifepristone on apoptosis in human uterine leiomyoma in vitro.Methods:Human uterine leiomyoma cells in vitro were treated with mifepristone at different concentrations and then cultured for 48 hours, the cell cycle and apoptotic percentage were detected by flow cytometry.Results:Mifepristone at different concentrations could induce apoptosis of human uterine leiomyoma in vitro, the apoptotic rate was increased with the increase of concentration of mifepristone (P<0.05).The ratio of G_2/M phase cells was reduced at lower concentration of mifepristone (10~ -8 mol/L), and both of ratio of S and G_2/M phase cells was reduced at higher concentrations (10~ -4 mol/L) (P<0.05 and P<0.01).Conclusion:Mifepristone can induce the apoptosis in human uterine leiomyoma in vitro, and there is an accordant concentration-response relationship, lower and higher concentrations of mifepristone could induce possibly the apoptosis of human uterine leiomyoma cells in different phase.
