摘 要: | 目的:了解单纯疱疹病毒(HSV)共有型单克隆抗体5B4-6的体内被动保护能力。方法:本研究分别采用体外中和实验和体内被动保护实验以检测单克隆抗体5B4-6的体外中和HSV的能力和体内被动保护BALB/C小鼠于足垫抗受HSV致死剂量攻击的保护能力。结果:1.单抗腹水5B4-6具有HSV-1及HSV-2交叉中和性,但对HSV-1的中和效应要强一些,其中和效价各为1:80(豚鼠新鲜血清(GPS)存在,HSV-1)、1:20(GPS缺乏,HSV-1)及1:20(GPS存在,HSV-2)、1:5(GPS缺乏,HSV-2)。单抗6B3-2-5仅具有抗HSV-1特异性的中和作用,其中和效价各为1:80(GPS存在)及1:20(GPS缺乏),而对HSV-2在GPS存在时才具有极低滴度的中和作用(1:5)。从中可发现补体参与其中和过程,但不是完全依赖。2.体内被动保护实验表明,单抗5B4-6具有极明显的保护小鼠于足垫接受HSV-1及HSV-2致死剂量攻击的保护能力,其保护率各为100%及80%。结论:单克隆抗体5B4-6具有一定的体内被动保护能力。

Protection Against Lethal Challenge in BALB/C Mice by Passive Transfer of a Type-common Monoclonal Antibody to Herpes Simplex Virus |

Abstract: | Objective:To investigate the protective effect of a type common monoclonal antibody 5B 4 6 against HSV in vivo.Methods:The neutralization activity to HSV in vitro and the protective activity against the lethal challenge of HSV of monoclonal antibody 5B 4 6 in BALB/C mice were detected by neutralization test and passive transfer test respectively.Results:1.Neutralization test in vitro showed that monoclonal antibody 5B 4 6 had a cross reactive neutralization activity to HSV 1 and HSV 2, the neutralization tiers to HSV 1 was significantly higher than that to HSV 2 and the neutralization titers in the presence of complements (fresh Guinea pig serum) significantly higher than that in the absence of complements.Monoclonal antibody 6B 3 2 5 showed neutralization activity only to HSV 1 with the titers of 1:80(in the presence of complements) and 1:20 (in the absence of complements).These results indicated that complements participated in the viral neutralization,but was not fully dependent .2.Protection against lethal HSV challenge in BALB/C mice could be achieved by passive transfer of monoclonal antibody 5B 4 6,with the protection rates of 100% and 80% to HSV 1 and HSV 2 respectively.Conclusion:monoclonal antibody 5B 4 6 has protective activity to some extent against the challege of HSV. |
