加味当归补血汤对肿瘤化疗所致小鼠免疫功能低下的影响

目的研究加味补血汤对荷瘤小鼠进行环磷酰胺(CY)化疗所致小鼠免疫功能低下的正向调节作用,筛选最佳治疗剂量。方法采用全自动血象分析仪检测RBC、WBC水平,采用MTT法测定淋巴细胞增殖程度、酶联免疫吸附法检测G-CSF活性观察加味当归补血汤对荷瘤小鼠CY化疗后免疫功能抑制的调节作用。结果 1)肿瘤组各项检测指标均下降,与空白组相比较有显著性差异(P<0.01或P<0.05);2)荷瘤小鼠腹腔注射CY后与肿瘤组小鼠相比较差异有统计学意义(P<0.01或P<0.05);3)各给药组小鼠的RBC、WBC、Hb水平G-CSF活性及T淋巴细胞增殖程度与模型组相比较,大部分存在差异有统计学意义(P<0.01或P<0.05),其中加味当归补血汤的中剂量治疗效果更加显著(P<0.01)。结论加味当归补血汤对肿瘤化疗所致小鼠免疫功能低下有上调作用,能有效的抑制CY导致免疫功能低下,中剂量效果最佳。

Experimental studies on the effect of Modified Dangguibuxue Decoction on immune function of mice induced by chemotherapy

Objective To explore the effect of positive regulation of Modified Dangguibuxue Decoction on immune function in tumor-bearing mice after chemotherapy with cyclophosphamide and screen optimal therapeutic dose. Methods Automatic blood analyzer was used to detect the levels of RBC and WBC. MTT assay was determined by the degree of lymphocyte proliferation. Enzyme-linked immunosorbent assay was used to detect the activity of G-CSF. Results： 1） The detection of indicators in tumor group decreased significantly compared with the that of the control group（ P〈0.01 或 P〈0.05）;2） Tumor-bearing mice after intraperitoneal injection of CY were statistically different from tumor mice（ P〈0. 01 或 P〈0. 05）; 3） Each administered group of mice was statistically different compared with the model group on RBC,WBC,G-CSF levels and the degree of T lymphocyte proliferation（ P〈0. 01 或 P〈0. 05）,treatment effect of Medium dose is more significant（ P〈0. 01）. Conclusion Modified Dangguibuxue Decoction raised immune function of chemotherapy-induced mice and effectively inhibited CY lead to immune dysfunction,and the medium dose is the best.
