摘    要:目的:建立简便易用的PON2基因单核苷酸多态(rs12026)的检测方法。方法:应用创造酶切位点原理设计引物,通过PCR-RFLP技术判断PON2基因SNP位点多态性。结果:设计一对特异引物并PCR扩增含PON2多态位点的DNA片段,根据限制性内切酶BsmAI酶切结果判断PON2位点多态性,PCR和酶切效果均较好。结论:应用创造酶切位点PCR-RFLP原理建立的PON2多态位点的检测方法具备简便、经济、快速的特点。

Abstract:Objective: To develop a proper assay for identifying single nucleotide polymorphism(SNP) of PON2 gene.Methods: PCR primers were designed by create restriction site(CRS) method,then PCR-RFLP was adopted to identify the SNP in PON2 gene.Results: One primer pair can get target products containing PON2 SNP site by PCR,restriction enzymes BSMAI were adopted to identify the SNP site.The results of PCR and RFLP were good.Conclusion: It suggested that the method of detecting the SNP of PON2 by CRS-PCR-RFLP theory was facility,economy,and rapid.
