鼻型NK／T细胞淋巴瘤中Survivin和caspase-3表达意义的初步研究

目的：探讨Survivin和caspase-3在鼻型NK／T细胞淋巴瘤中的表达，并初步分析其在鼻型NK／T细胞淋巴瘤发生发展中的意义。方法：应用免疫组化EnVision二步法检测50例鼻型NK／T细胞淋巴瘤和12例正常淋巴结组织中Survivin和caspase-3的表达水平。结果：50例肿瘤组织中Survivin阳性表达率为60．0％（30／50），12例正常淋巴结组织中Survivin阳性表达率为16．7％（2／12），且主要局限于少数生发中心细胞（χ^2=7．28，P〈0．05）。caspase-3在肿瘤组织和正常淋巴结中的阳性表达率分别为84．0％（42／50）和100．0％（12／12），两者无显著性差异（χ^2=2．2，P〉0．05）。Survivin基因高表达与caspase-3基因表达下调密切相关（χ^2=4．86，P〈0．05），Survivin的表达与caspase-3的表达呈负相关（r=-0．953，P〈0．05）。结论：Survivin高表达在鼻型NK／T细胞淋巴瘤的发生发展中可能有一定的作用。Survivin与caspase-3的表达呈负相关，Survivin可能通过抑制caspase-3的激活而发挥其抑制细胞凋亡的作用。

Expression of survivin and caspase-3 in nasal-NK/T cell lymphoma and its significance

Objective:To investigate the expression of survivin and caspase-3 in NK/T cell lymphoma and its significance. Methods:Immunohistochemistry EnVision system was applied to test the expression of survivin and caspase-3 in 50 cases of nasal-NK/T cell lymphoma and 12 cases of normal lymph nodes. Results:The positive expression rates of survivin in tumor tissues and nomal lymph nodes were 60.0%(30/50) and 16.7%(2/11), respectively, and there was significant difference between them(P<0.05). The positive expression of caspase-3 was 100% in nomal lymph nodes, which was higher than that in tumor tissues, but there was no significant difference (P>0.05). Negative correlation between the expression of survivin of caspase-3 was found. Conclusion:Survivin may play an important role in the carcinogenesis of NK/T cell lymphomas by inhibiting cell apoptosis and promoting cell proliferating. Moreover survivin may prevent cell apoptosis by inhibiting the activity of caspase-3.
