大鼠C6脑胶质瘤CT灌注成像的初步研究

摘    要:目的初步探讨CT灌注成像技术用于大鼠C6脑胶质瘤微循环研究的可行性。方法采用立体定向方法在20只雌性Wistar大鼠右侧尾状核接种C6细胞,荷瘤大鼠分为二组,一组于接种后1、2、3周分别行MR检查以评价肿瘤生长,另一组于接种后2周时行CT灌注成像,每次检查结束后留取鼠脑标本作病理学检查。结果大鼠C6脑胶质瘤的生长特征及MR表现符合文献报道。肿瘤区脑血流量(CBF)、脑血容量(CBV)、对比剂平均通过时间(MTT)和表面通透性(PS)及瘤脑交界处CBV,MTT,PS值均明显大于对侧正常脑组织(P<0.05),肿瘤中心PS值大于肿瘤周边(P<0.05),CBF、CBV、MTT则无统计学差异(P>0.05);肿瘤区有较多新生血管,肿瘤周边更为丰富。结论CT灌注成像可用于研究大鼠C6脑胶质瘤的微循环变化。

Preliminary study of CT perfusion imaging in rat C6 brain gliomas

Abstract:Objective To study the feasibility of CT perfusion in rat C6 brain gliomas. Methods C6 cells were inoculated into the right caudate nucleus of 20 Wistar rats by stereotactic procedure to established animal model. Some tumor-bearing rats were used to observe tumor development by MRI, which was performed on 1, 2, 3 weeks after implantation, others were used to study CT perfusion on 2 weeks after implantation. Immediately after each examination, some rats were sacrificed for pathology. Results The development characteristics of rat C6 brain gliomas on MRI and pathology were consistent with previous published reports. The value of cerebral blood flow (CBF), cerebral blood volume (CBV), mean transmit time (MTT), permeability surface (PS) in tumors except for CBF in tumor-brain areas increased significantly compared with that on the contralateral brain (P<0.05). Then the value of PS in the central areas of the tumors was higher than that in the peripheral areas while there was no difference of other parameters between the areas. Furthermore, there was more angiogenesis in the tumors than that on the contralateral brain especially in the periphery of the tumors. Conclusion CT perfusion imaging can be used to study the microcirculation of rat C6 brain gliomas.
