山羊下颌骨牵张成骨过程中间隙连接蛋白43的表达

摘    要:目的:探讨牵张成骨过程中连接蛋白43(Cx43)调节成骨细胞的骨形成功能与破骨细胞的骨吸收功能之间的偶联动态平衡的作用机制。方法:在建立山羊下颌骨牵张成骨动物模型的基础上,应用免疫组化法观察牵张成骨不同时期骨组织中Cx43的表达以及分布情况。计算骨组织细胞中Cx43的阳性表达率.应用SigmaStat2.03软件包进行单因素方差分析。结果:Cx43表达主要位于成骨细胞、破骨细胞和骨细胞的细胞膜上。牵张1周后,牵张区成骨细胞、破骨细胞Cx43阳性表达率分别为(59.7±3.0)%和(56.8±4.0)%,两者无显著差异(P〉0.05)。骨细胞Cx43阳性表达率为(29.0±2.8)%,显著低于成骨细胞和破骨细胞(P〈0.05)。牵张2周后,牵张区的成骨细胞、破骨细胞和骨细胞的Cx43的阳性表达率分别为(56.7±5.3)%、(49.0±4.1)%和(36.2±5.1)%,成骨细胞Cx43的阳性表达率显著高于破骨细胞和骨细胞(P〈0.05),表达主要位于细胞膜上,胞质中亦有少许表达。完成牵张4周及6周后,新骨形成区成骨细胞、破骨细胞和骨细胞细胞膜上均有Cx43表达,其阳性表达率无显著差异(P〉0.05)。结论:牵张成骨过程中,Cx43对成骨细胞的骨形成功能与破骨细胞的骨吸收功能之间的偶联动态平衡有比较重要的调节作用。

Abstract:PURPOSE: To study the effect of Connexin 43(Cx43) on the interaction communication between bone-forming osteoblasts and bone-absorbing osteoclasts during distraction osteogenesis. METHODS: The animal model of goat mandibular distraction osteogenesis was established. The expression location of Cx43 in the bone tissue was determined with immunohistochemical assay during different distraction phase. Positive rates of Cx43 were measured and the data was statistically analyzed with One-way ANOVA using SigmaStat2.03 Software package. RESULTS: The expression of Cx43 was mainly localized in cell membrane of osteoblasts,osteoclasts and osteocytes. The positive rates of Cx43 in osteocytes (29.0±2.8)% was significantly lower than that in osteoblasts (59.7±3.0)% and osteoclasts (56.8±4.0)% one week after distraction (P〈0.05). Osteoblasts indicated significantly higher positive rates of Cx43 (56.7±5.3)% when compared to osteoclasts (49.0±4.1)% and osteocytes (36.2±5.1)% 2 weeks after distraction (P〈0.05). Four weeks and 6 weeks after distraction, no significant difference of Cx43 positive rates was observed among osteoblasts, osteoclasts and osteocytes (P〉0.05). CONCLUSION: Cx43 possibly plays a role in the coupling regulation between bone-forming osteoblasts and bone-absorbing osteoclasts during distraction osteogenesis.
