| 摘 要： | 目的：研究体外胚肺成纤维细胞对肺癌H460细胞增殖及MMP-2的影响。方法：在单层平面培养条件下,H460细胞和与胚肺成纤维细胞条件培养基共同培养;单层平面及三维立体培养条件下,H460细胞和胚肺成纤维细胞共同培养,MTT或明胶酶谱法测定H460细胞的增殖活力及MMP-2的表达。结果：胚肺成纤维细胞条件培养基不能促进肺癌H460细胞的增殖和MMP-2的表达。胚肺成纤维细胞与肺癌H460细胞共同培养时出现了MMP-2的表达明显增强。结论：胚肺成纤维细胞能通过促进H460细胞MMP-2的表达影响肺癌的侵袭和转移。

| Abstract: | Objective：To investigate the effection of fetal lung fibroblasts on the MMP-2 expression and proliferation of lung cancer cells in vitro.Methods：H460 were cultured in fetal lung fibroblasts-conditioned media,with or without fetal lung fibroblasts in monolayer or in three dimensional culture models.MTT assay and gelatin zymography were used to examine the cell proliferation and MMP-2 expression.Results：The conditioned media had no effect on the proliferation of H460 cells and MMP-2 expression.The expression of MMP-2 was greatly enhanced when the cells were co-cultured with fetal lung fibroblasts.Conclusion：Fetal lung fibroblasts can influence the invasion and metastasis of lung cancer cell H460 by enhancing the expression of MMP-2. |
