摘    要:目的 观察日本血吸虫未成熟虫卵超免疫血清对虫卵胚胎发育及雌虫生殖力影响的效果。方法 本研究通过体外培养系统 ,观察日本血吸虫成虫离体产卵及虫卵发育全过程中形态学特点的变化 ,检测抗未成熟虫卵超免疫血清对虫卵胚胎发育及雌虫生殖力的影响。结果 在含正常兔血清的培养基中 ,初产卵经 12~ 14天培体外培养后 ,能完成发育至成熟期 ,对照组雌虫每周产卵总数为 176 5 7个 (n =2 0 )。但在含有抗未成熟虫卵超免疫兔血清的培养基中 ,雌虫产卵数量较正常兔血清培养对照组有显著降低 (P <0 0 0 1) ;在培养后第 10天 ,试验组雌虫即停止产卵 ,部分初产卵卵胚的发育和成熟过程被完全阻断 ;免疫荧光染色显示 ,抗卵免疫血清可作用于卵胚细胞及雌虫卵黄腺、肠腔内膜组织。结论 体外培养结果表明 ,日本血吸虫未成熟虫卵抗原诱导的免疫应答 ,在抗虫卵胚胎发育及抗雌虫生殖产卵中起重要作用。

Abstract:In this study,the extra-host oviposition of the adult worms of Schistosoma japonicum and the whole process of egg development were observed by means of in vitro cultural system.Eggs laid in the same period by the in vitro cultured female worms were cultivated and their morphological characteristics in the developmental process were observed.Effects of the hyperimmune serum against S.japonicum immature egg antigen (SIEA) on the embryo development and the fecundity of female worms were assayed.The results showed that the number of eggs laid by the adult worms cultivated with the SIEA hyperimmune serum was significantly lower than that in the control group.Adult worms in the experimental group started to stop laying eggs from the 10 th day after cultivation.The embryo development and maturation of the newly laid eggs by the worms in the experimental group was markedly inhibited,and the number of denatured and undeveloped eggs in the experimental group was significantly increased than that in the control group.The immunofluorescence staining revealed that the SIEA hyperimmune serum could react with the vitellaria and the lining membrane tissues of the gut lumen of female worms as well as with the embryo cells.These results suggest that SIEA may play a role in the immune responses against the egg development and fecundity of S.japonicum.
