七氟烷麻醉对发育期大鼠神经功能及认知功能影响

摘    要:目的探讨新生期大鼠吸入七氟烷对其神经及认知功能发育的影响作用及其机制。方法选取新生SD大鼠48只,采用随机数字表法分为实验组和对照组各24只,实验组分别于出生后第7、14、21d吸入2.6%七氟烷+运载气体2h,对照组吸入正常空气;对比两组大鼠喂实验第30d时(出生后37d),对比Morris水迷宫实验中游泳路程、逃避潜伏期、原平台象限滞留时间、原平台象限游泳距离,采用Western-blot技术检测两组大鼠海马组织中脑源性神经营养因子(BDNF)、突触后致密物质95(PSD-95)、Tau蛋白的表达差异;采用ELISA法检测各组大鼠脑脊液中载脂蛋白E(ApoE)水平。结果在出生后37d测定,实验组和对照组大鼠的Morris水迷宫实验中游泳路程、原平台象限游泳距离差异无统计学意义(P>0.05),实验组大鼠的Morris水迷宫实验中逃避潜伏期、原平台象限滞留时间均较对照组长(P<0.05);实验组大鼠的海马组织中BDNF、PSD-95蛋白表达低于对照组(P<0.05),Tau蛋白表达强度高于对照组(P<0.05);实验组大鼠脑脊液中ApoE水平高于对照组(P<0.05)。结论新生期大鼠吸入七氟烷对其神经及认知功能发育在早期有一定的影响,可能与影响海马组织中BDNF、PSD-95、Tau蛋白表达有关。

Abstract:Objective To investigate the effects of inhaled sevoflurane on the development of neurological and cognitive functions in neonatal rats and its mechanism.Methods Forty-eight newborn SD rats were randomly divided into the experimental group and the control group.The experimental group inhaled 2.6%sevoflurane+carrier gas for 2 hours at 7th,14th and 21st after birth.Normal air;comparing the two groups of rats on the 30th day of the experiment(37 days after birth),comparing the swimming distance,escape latency,original platform quadrant retention time,and original platform quadrant swimming distance in the Morris water maze experiment,using Western-blot technique to detect two The expression of brain-derived neurotrophic factor(BDNF),post-synaptic dense substance 95(PSD-95)and Tau protein in hippocampus of rats was detected.The apolipoprotein E(ApoE)in cerebrospinal fluid of each group was detected by ELISA.)Level.Results There was no significant difference in the swimming distance and the original platform quadrant swimming distance between the experimental group and the control group at 37 days after birth(P>0.05).The escape latency of the Morris water maze test in the experimental group was observed.The retention time of the original platform quadrant was longer than that of the control group(P<0.05).The expression of BDNF and PSD-95 protein in the hippocampus of the experimental group was lower than that of the control group(P<0.05),and the expression of Tau protein was higher than that of the control group(P<0.05);the ApoE level in the cerebrospinal fluid of the experimental group was higher than that of the control group(P<0.05).Conclusion Inhalation of sevoflurane in neonatal rats has a certain effect on the development of neurological and cognitive functions in early stage,which may be related to the expression of BDNF,PSD-95 and Tau protein in hippocampus.
