地龙耐热蛋白对自发性高血压大鼠心肌血管紧张素Ⅱ水平及血管紧张素Ⅱ-1型受体表达的影响

摘    要:目的探讨地龙耐热蛋白对自发性高血压大鼠(SHR)心肌血管紧张素Ⅱ(AngⅡ)水平及AngⅡ-1型(AT1)受体表达的影响。方法采用放射免疫法测定血浆和心肌AngⅡ水平。采用免疫荧光法测定心肌AT1受体表达水平。结果模型组血浆和心肌AngⅡ水平均显著高于对照组(P<0.01),地龙耐热蛋白高剂量组、低剂量组血浆和心肌AngⅡ水平均低于模型组(P<0.05)。对照组与模型组比较,心肌细胞膜和胞浆AT1受体表达减少;地龙耐热蛋白低剂量组和高剂量组与模型组比较,心肌细胞膜和胞浆AT1受体表达均显著减少。结论地龙耐热蛋白可降低血浆、心肌AngⅡ水平,下调心肌AT1受体的表达,且其下调机制可能与降低心肌AngⅡ水平有关。

Abstract:Objective It is to discuss the influence of earth worm thermostable protein(EWTP) on myocardial angiotensin Ⅱ(AngⅡ) level and angiotensin Ⅱ receptor-1(AT1) expression of spontaneously hypertensive rat(SHR).Methods Plasma and myocardial AngⅡ levels were detected with radioimmunoassay.The expression level of myocardial AT1 receptor was detected with immunofluorescence.Results Plasma and myocardial AngⅡ levels in model group were significantly higher than those in control group(P<0.01).Plasma and mycardical AngⅡ levels in EWTP high dosage group and low dosage group were lower than those in model group(P<0.05).Compared with model group,the expression of AT1 receptor in myocardial cytolemma and endochylema in control group was decreased.Compared with model group,the expression of AT1 receptor in myocardial cytolemma and endochylema in EWTP high dosage group and low dosage group was significantly decreased.Conclusion EWTP can lower plasma and myocardial AngⅡ levels and can down regulate myocardial AT1 receptor expression.Its down regulating mechanism was probably related to lowering myocardial AngⅡ level.
