宫颈癌中PTEN、p16、p27的表达及临床意义

摘    要:目的:研究宫颈癌组织中肿瘤抑制基因PTEN、p16、p27的表达,探讨它们在宫颈癌诊断、治疗中的价值。方法:应用免疫组化技术,分别检测正常宫颈组织和宫颈癌组织中PTEN、p16、p273种肿瘤抑制基因蛋白表达,结合临床病例的分期、肿瘤分级、淋巴结是否转移进行统计学分析。结果:正常宫颈组织中PTEN蛋白、p16蛋白及p27蛋白的表达率分别是95%、20%和100%。PTEN、p27的表达下降与宫颈癌的病理分型无明显关系,与宫颈癌的临床分期、组织学分级及有无淋巴结转移关系密切。p16在宫颈癌组织中呈现过表达,但过表达与病理分型、临床分期、组织学分级及有无淋巴结转移无明显关系。结论:PTEN蛋白、p27蛋白在宫颈癌进展中起重要作用;在宫颈癌组织中p16蛋白表现为过表达,是宫颈癌的一类早发事件。在宫颈癌组织中PTEN蛋白与p27蛋白表达强度之间存在正相关性(P<0.05)。

Abstract:Objective:To study the relationship between tumor suppressor gene PTEN,p16,p27 and primary cervical carcinomas.Methods:The expresssion of PTEN,pl6 and p27 in normal cervical tissues and primary cervical carcinomas were detected by immunohistochemistry.Results:The positive expression rate of PTEN protein was 95% in normal cervical tissues,the rate of pl6 protein was 20% and the rate of p27protein was 100%. Gene PTEN,p27 was not associated with clinical pathologic parameters but expression of PTEN,P27 protein was irrelevant to the stage,grade and metastasis of lymph nodes. The loss of pl6 protein was not associated with cervical carcinomas,but the high expression of pl6 was found in the cervical carcinomas,the rate was not irrelevant to the clinical pathologic parameters,stage,grade and metastasis of lymph nodes,p16 was found early in the cervical carcinomas,p27 and PTEN showed significantly positive correlation in cervical carcinomas (P<0.05).Conclusion:The expression of PTEN and p27 protein play an important role in the development of uterine cervical neoplasms,p27 high-level expression in uterine cervical neoplasms.
