IL-8和CXCL12在来曲唑诱导的PCOS大鼠卵巢中的表达及意义

目的:应用来曲唑建造多囊卵巢综合征(PCOS)大鼠模型,探讨趋化因白介素-8和CXCL12在PCOS发病机制中的作用。方法:应用来曲唑诱导PCOS大鼠模型,阴道涂片观察大鼠动情周期变化,应用放免法测定大鼠血清性激素水平;大鼠卵巢组织石蜡包埋、组织切片并HE染色观察组织学变化;借助免疫组织化学技术,对IL-8和CXCL12在多囊卵巢综合征大鼠和对照组大鼠卵巢组织的表达情况进行对照研究。结果:PCOS大鼠模型建造成功,PCOS组IL-8和CXCL12在卵泡膜细胞、颗粒细胞、间质细胞的表达明显低于实验对照组(P<0.05)和空白对照组(P<0.05)。结论:①来曲唑诱导的大鼠动物模型是理想的PCOS模型。②IL-8和CXCL12参与多囊卵巢综合征排卵障碍的发生。

Expression and significance of interleukin-8 and CXCL12 in ovarian of polycystic ovary syndrome induced by letrozole in rats

Objective:To establish an animal model of polycystic ovarian syndrome(PCOS) by letrozole in rats,explore the effect of interleukin-8(IL-8) and chemokine ligand 12(CXCL12) in the pathogenesis of PCOS.Methods:Changes of estrus in rats were observed by vaginal smear everyday.The levels of gonadal hormones in serum were detected by radioimmunoassay(RIA).Histologic changes in ovaries were observed by paraffin imbedding,tissue slice and HE staining.Expression levels of IL-8 and CXCL12 of ovary in PCOS rats,control group and blank group were detected by immunohistochemical staining.Results:The levels of IL-8 and CXCL12 of PCOS rats in follicles theca cells,granulosa cells,stromal cells were significantly lower than those of control group and blank group(P<0.05).Conclusion:Letrozole-induced model in rats is an ideal model of PCOS.IL-8 and CXCL12 may have significant effect on the pathogenesy of PCOS.
