选择性雌激素受体调节剂对放疗所致卵巢早衰引起骨质丢失的实验研究

摘    要:目的:通过观察选择性雌激素受体调节剂(雷洛昔芬)对放疗去势大鼠的骨代谢生化指标的影响,探讨选择性雌激素受体调节剂对放疗所致卵巢早衰引起骨质丢失的保护作用。方法:选用3月龄雌性wistar大鼠40只,随机分为4组:假放疗组、放疗组、放疗+雷洛昔芬组、放疗+己烯雌酚组,每组10只,16周后,处死大鼠,腹主动脉取血,检测血清雌二醇、卵泡刺激素,骨钙素及碱性磷酸酶。结果:放疗组及放疗+雷洛昔芬组雌二醇水平明显低于假放疗组,差异有显著意义(P<0.05),而卵泡刺激素水平明显高于假放疗组(P<0.05);放疗组骨钙素和碱性磷酸酶明显高于其他组,差异有显著意义,而另3组间差异不显著。结论:大鼠卵巢放疗可致去势;放疗去势后大鼠有骨质丢失,雷洛昔芬对放疗去势后大鼠的骨丢失和己烯雌酚一样有保护作用。

Abstract:Objective:To explore the protective effect of selective estrogen receptor modulators(SERMs) on bone metabolism induced by premature ovarian failure(POF) after radiotherapy in rats.Methods:40 female Wistar rats aged 3 months were randomly divided into 4 groups(10 rats in every group):Group A(sham radiotherapy),Group B(radiotherapy),Group C(radiotherapy and raloxifene),Group D(radiotherapy and diethylstilbestrol).All rats were killed 16 weeks later,then levels of estradiol(E2),follicle stimulating hormone(FSH),bone gla protein(BGP) and alkaline phosphatase(ALP) in serum were detected.Results:The levels of E2 in group B and group C were lower than that in group A(P<0.05),while the levels of FSH in group B and group C were higher than that in group A(P<0.05);the levels of BGP and ALP in group B were higher than those in the other groups(P<0.05),but there was no significant difference between group A,group C and group D.Conclusion:Rats will be gonadectomy after radiotherapy to ovaries,then bone loss is found in those rats.Rloxifene has protective effect on bone loss of rats induced by gonadectomy after radiotherapy,which is alike to diethylstilbestrol.
