毛冬青总黄酮对小鼠脑缺血模型的影响

摘    要:目的探讨毛冬青总黄酮对小鼠急性脑缺血模型和小鼠短暂脑缺血再灌注模型的影响。方法造模组小鼠连续给药10 d,于最后1次给药后1 h,采用迅速断头法造成急性脑缺血模型,记录小鼠的呼吸次数及呼吸维持时间;采用阻断小鼠双侧颈总动脉10 min,再灌注15 min,造成小鼠短暂性脑缺血再灌注模型,立即测定凝血时间。分离脑,一半脑置于10%福尔马林溶液中固定,做海马区病理切片;另一半脑制备成10%的脑匀浆,用于测定ATP酶(Na+-K+-ATPase、Ca++-Mg++-ATPase、Ca++-ATPase、Mg++-ATPase)和乳酸(LD)含量。结果毛冬青总黄酮能显著增加急性脑缺血模型小鼠的呼吸次数、延长呼吸时间;还能明显延长小鼠短暂性脑缺血再灌注模型的凝血时间,明显提高脑匀浆中Na+-K+-ATPase、Ca++-Mg++-ATPase、Ca++-ATPase、Mg++-AT-Pase活力;显著降低脑匀浆中LD含量;显著改善脑组织病理改变。结论毛冬青总黄酮可显著改善小鼠急性脑缺血模型和短暂脑缺血再灌注模型的脑缺血状况,具有脑保护作用。

Abstract:Objective To investigate the effect the total flavonoids of Radix Ilicis Pubescentis in mice with acute cerebral ischemia and transient cerebral ischemia and reperfusion.Methods The experimental mice were fed with total flavonoids of Radix Ilicis Pubescentis for ten continuously days.One hour after the last medication,we established the model of acute cerebral ischemia by rapid head-breaking method,and recorded the frequency and duration of breathing.By the method of occlusion of bilateral common carotid artery for 10 min and reperfusion for 10 min,we established the model of transient cerebral ischemia,and detected clotting time immediately.After isolation of the brain,we put half of the brain in 10 % formalin solution for HE staining,and he other half was prepared into brain homogenate for the determination of ATPase(Na+-K+-ATPase,Ca++-Mg++-ATPase,Ca++-ATPase,Mg++-ATPase)and lactic acid.Results The total flavonoids of Radix Ilicis Pubescentis significantly increased respiratory rate and prolonged breathing time in mice after head breaking.In mice models of transient cerebral ischemia-reperfusion,clotting time was prolonged significantly,Na+-K+-ATPase,Ca++-Mg++-ATPase,Ca++-ATPase,and Mg++-ATPase activity in the brain homogenate were increased significantly,content of LD in brain homogenate was decreased significantly,and pathological changes in the brain tissue were improved significantly.Conclusion The total flavonoids of Radix Ilicis Pubescentis show protective effect on improving cerebral ischemic conditions of cerebral ischemia mice.
