摘    要:目的研究体外不同低氧分压对SD大鼠骨髓间充质干细胞(MSCs)增殖分化的影响。方法将体外培养的MSCs,分组置于常氧(21%)和低氧(2%、4%、6%、8%)培养箱中培养,用MTT法分别于1、3、5、7、9d检测细胞的增殖活性;用酶联免疫检测法分别于1、3、5、7、9d检测细胞碱性磷酸酶(ALP)和Ⅰ型胶原蛋白(COLⅠ)的表达。结果不同低氧分压对MSCs细胞增殖活性有明显促进作用(P<0.05);低氧分压抑制MSCs细胞的ALP和COLⅠ表达(P<0.05)。结论低氧微环境可促进MSCs的增殖活性,但对细胞的成骨向分化起到抑制作用。

Abstract:Objective To investigate the effects of different low partial pressure of oxygen on proliferation and differentiation capability of rat mesenchymal stem cells(MSCs) in vitro.Methods MSCs were isolated from adult rat bone marrow and cultured in vitro under normoxia(21% oxygen tension) and low PO2(2%,4%,6% and 8% oxygen tension) respectively.The proliferation of MSCs,alkaline phosphatase(ALP) activity,and production of collagens type Ⅰ(COLⅠ) were examined by means of MTT and enzyme-linked immunoreaction on1,3,5,7,9 d respectively.Results Different low partial oxygen pressures had significant promoting effect on the proliferation activity of MSCs(P<0.05) and decreased ALP activity and the production of COLⅠ(P<0.05).Conclusion Hypoxia microenvironment can promote the proliferation activity of MSCs and inhibit MSCs to differentiate into osteogenic lineage cells.
