参芪化淤方对H22肝癌细胞survivin表达的影响

摘    要:目的探讨参芪化淤方对H22肝癌细胞survivin基因表达的影响。方法将H22肝癌荷瘤小鼠60只随机分为6组,参芪化淤方高、中、低剂量组,环磷酰胺(CTX)对照组,模型组,阴性对照组。参芪化淤方低、中、高剂量组分别予以0.4mL参芪化淤方(低剂量组:1.04g/mL,中剂量组:2.08g/mL,高剂量组:4.16g/mL)灌胃,CTX对照组予以0.2mL CTX溶液(小鼠给药剂量为20mg.kg-1.d-1)腹腔注射,模型组予以0.4mL生理盐水灌胃,每日1次,给药14d。免疫组化法检测肿瘤组织sur-vivin基因的表达;同时观察脾、胸腺指数及肿瘤抑制率。结果参芪化淤方高、中、低剂量对小鼠的脾及胸腺均具有一定保护作用,能升高小鼠的脾、胸腺指数。其中低剂量组与CTX对照组对脾及胸腺的保护作用相当,与模型组比较,差异有统计学意义(P<0.05),而中、高剂量组对脾及胸腺的保护作用更强(P<0.01)。与模型组比较,参芪化淤方中、高剂量组对H22型肝癌小鼠肿瘤抑制作用明显(P<0.01),低剂量组和CTX对照组也具有一定的抑制作用(P<0.05)。参芪化淤方高、中、低剂量组都能降低H22肝癌荷瘤小鼠癌组织中survivin基因的表达水平,其中CTX对照组及低剂量组与模型组比较差异有统计学意义(P<0.05),中、高剂量组的抑制作用更显著(P<0.01)。结论参芪化淤方对H22肝癌荷瘤小鼠癌组织中survivin基因的表达具有一定抑制作用,且能保护小鼠的免疫器官。

Abstract:Objective To study the effects of Shenqihuayu formula on the survivin gene expression of H22 hepatoma cells.Methods Mice bearing H22 hepatoma cells were randomly divided into 6 groups:high dose,medium dose and low dose of Shenqihuayu formula,CTX group,model group and negative control group.The high,medium and low dose groups were given 0.4 g/mL,1.04 g/mL,2.08 g/mL of Shenqihuayu formula by gastric gavage.The CTX group was intraperitoneally given CTX solution 0.2 mL(dose of 20 mg·kg-1·d-1) and the model group was given normal saline 0.4 mL,once a day for 14 d.The expression of survivin gene of tumor tissues was tested by the immunohistochemical method.At the same time,the spleen,thymus index and tumor inhibition rate were observed.Results Low-dose,middle-dose and high-dose of Shenqihuayu formula all had some protective effects on the spleen and thymus of mice andincresed the spleen and thymus indexes.Among them,the protective effects on spleen and thymus in the low-dose group and the CTX control group were similar,but showing statistical difference compared with the model group(P<0.05).The protective effects in the middle-dose and high-dose groups were stronger(P<0.01).Compared with the model group,the inhibition effects on mice′ s H22 hepatoma cells in the middle-dose and high-dose groups were obvious(P<0.01),the low-dose group and the CTX control group also showed certain inhibition effects(P<0.05).All dose groups of Shenqihuayu formula could reduce the expression of survivin gene.Comparison between the CTX control group and the low-dose group with the model group showed statistical difference(P<0.05),the middle-dose and high-dose groups had more obvious inhibition effect(P<0.01).Conclusion Shenqihuayu formula can inhibit the expression of survivin to some extent and protect the immune organs of mice with H22 hepatoma cells.
