IL-10对脑出血大鼠HSP70、caspase-3表达的影响

摘    要:目的观察白细胞介素10(IL-10)对出血性颅脑损伤大鼠脑组织热休克蛋白70(HSP70)、胱天蛋白酶3(caspase-3)表达的影响,以探讨IL-10对出血性颅脑损伤的保护作用及机制。方法 96只SD大鼠随机分为正常组(N组)、模型组(M组)、IL-10低剂量组(L组)、IL-10高剂量组(H组)。Ⅶ型胶原酶注射法制备颅脑损伤模型,记录各组6h、12h、1d、3d、7d大鼠神经功能改善程度;免疫组化法测定HSP70和caspase-3的表达。结果与M组比较,L组和H组HSP70表达明显升高(P<0.01),caspase-3表达明显降低(P<0.01),神经功能缺损评分明显下降(P<0.01),且以H组更为显著(P<0.05)。结论 IL-10可通过上调HSP70表达及抑制caspase-3表达对出血性颅脑损伤起保护作用。

Abstract:Objective To observe the effects of IL-10 on expression of HSP70 and caspase-3 after intracerebral hemorrhage in rats and study the neuroprotection mechanism of IL-10 for intracerebral hemorrhage.Methods 96 SD rats were randomly divided into four groups:normal group(group N),model group(group M),Low-dose IL-10 treatment group(group L) and high-dose IL-10 treatment group(group H).The intracerebral hemorrhage rat model was established by intracranial injection of Ⅶ collagenase.The recovery of rats’ neurological function of all groups was recorded after 6 h,12 h,1 d,3 d,7 d;The expressions of HSP70 and caspase-3 were analyzed by immunohistochemistry.Results The expression of HSP70 in perihematoma in group L and H were upregulated(P<0.01),caspase-3 and the score of neurologic impairment were downregulated as compared to M group(P<0.01).The above roles of IL-10 were more significant in group H(P<0.05).Conclusion IL-10 plays protective role in hemorrhagic brain damage by upregulation of HSP70 and downregulation of caspases-3.
